痰热清注射液有效成分在大鼠肺组织靶向性研究 被引量:6

摘要 目的:通过清热化痰法的代表方痰热清注射液有效成分在大鼠肺组织的靶向性研究,初步探讨清热化痰法治疗慢性阻塞性肺疾病的机制。方法:运用高效液相色谱二级质谱联用方法对痰热清注射液制剂中的有效成分及其在大鼠血浆与组织中能够检测的有效成分进行定性定量分析和比较。结果:痰热清注射液制剂中能够检测出黄芩、金银花、连翘、熊胆粉4味配伍中药的有效成分,其中黄芩苷、绿原酸、咖啡酸、汉黄芩素、野黄芩苷能够进行定量检测。在大鼠血浆及肺组织中,能够同时定性定量分析痰热清注射液中首入肺经的黄芩、金银花两味中药的有效成分,其主要有效成分黄芩苷、绿原酸在肺组织的峰浓度明显高于其他组织。结论:大鼠血浆及肺组织中能够检测到清热化痰方剂痰热清注射液中的有效成分;痰热清注射液对肺组织具有一定的靶向性和中药归经作用;可通过方剂有效物质基础及组织靶向性的研究方式探讨清热化痰法治疗慢性阻塞性肺疾病的机制。 Objective: To primary explore the mechanism of Tanreqing injection,a representative formula of "Clearing Heat and Resolving Phlegm",in treatment of chronic obstructive pulmonary disease( COPD) by the effective components in vivo and the lung tissues. Methods: The effective components of Tanreqing injection was qualitatively and quantitatively determined by high performance liquid chromatography-tandem mass spectrometry( HPLC-MS / MS). The effective components of Tanreqing in the plasma and tissues of rats were qualitatively and quantitatively determined at different time points by HPLC-MS / MS after intravenous injection of Tanreqing. Results: A total of 10 effective components in Tanreqing were determined,including baicalin,chlorogenic acid,wogonin,scutellarin,caffeic acid,rutin,hyperin,phillyrin,ursodeoxycholic acid,chenodeoxycholic acid and baicalin,with chlorogenic acid,wogonin,scutellarin and caffeic acid quantitatively measured. The effective components of scutellaria and honeysuckle including baicalin,chlorogenic acid,wogonin,scutellarin,caffeic acid could be determined in the plasma and the tissue of rats and their concentration-time curve could be drawn up, which belonged to lungs in the traditional meridian tropism theory. The results showed that the effective components of Tanreqing could arrive at lung tissue with definite lung tissue targeting. Conclusion: The major effective components of Tanreqing could be measured in the plasma and tissues of rats. Tanreqing possessed definite lung tissues,which was in accordance with the traditional meridian tropism theory in lung. We could explore the mechanism of the method of "Clearing Heat and Resolving Phlegm" in treatment of COPD by the effective components of Tanreqing injection traditional in vivo and lung tissues.
