摘要
目的建立归及乳膏的质量标准。方法观察性状;以α-萘酚硫酸法鉴别总多糖;以薄层色谱法鉴别阿魏酸;以蒽酮染色法测定总多糖含量;以高效液相色谱法测定阿魏酸含量。结果归及乳膏为棕灰色的乳膏;α-萘酚硫酸鉴别呈总多糖的阳性反应;样品在硅胶G薄层板上与对照品相应位置显阿魏酸的紫色荧光斑点;多糖在1.875-33.75μg·m L^-1呈良好的线性关系(r=0.999 8),平均回收率均〉95%,RSD均〈1.2%,平均含量应为4.0 mg·g^-1;阿魏酸在3.0-96.0μg·m L^-1呈良好的线性关系(r=0.999 5),平均回收率〉95%,RSD均〈1.5%,平均含量应为60.0μg·g^-1。结论所建立质量标准可操作性强,结果重现性好,适用于归及乳膏的质量控制。
Objective To establish the quality standards of Guiji cream. Methods We observed the characters and identified the total polysaccharide by α-naphthol sulfuric acid method and the ferulic acid by thin layer chromatography(TLC). The total polysaccharide content was determined by anthrone staining and the determination of ferulic acid content by HPLC. Results Guiji cream was brown gray; α-naphthol sulfuric acid showed positive identification of polysaccharides. Silica gel TLC plate G, sample and ferulic acid reference substance showed purple fluorescent spots in the corresponding position. The linearity of polysaccharide was 1.875- 33.75 μg·m L^-1(r = 0.999 8), and the linearity of ferulic acid was 3.0- 96.0 μg·m L^- 1(r = 0.999 5). The average recoveries of polysaccharide and ferulic acid were more than 95%, and RSDs were less than 1.5%. The average content was 4.0 mg·g^- 1 for total polysaccharides, 60.0 μg·g^- 1for ferulic acid. Conclusion The method is accurate, reproducible and can be used for the quality control of Guiji cream.
