百合逍遥散对抑郁模型大鼠行为学及海马5-HT、NE的影响 被引量:12

Effects of Baihe Xiaoyao San on Behavior and Hippocampus 5-HT/NE of Depression Rats

摘要 目的:观察百合逍遥散对抑郁模型大鼠行为学及海马5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)水平的影响,探讨百合逍遥散对抑郁症的干预作用,并观察其与氟西汀是否具有协同抗抑郁作用。方法:60只雌性成年SD大鼠随机分为空白组、模型组、氟西汀组、百合逍遥散组及百合逍遥散联合氟西汀组(中西医结合组),各12只。除空白组外,各组均采用CUMS法造模28 d。造模成功后,氟西汀组给予1.8 mg·(kg·d)-1氟西汀,百合逍遥散组给予12.8 g·(kg·d)-1百合逍遥散,百合逍遥散联合氟西汀组予1.8 mg·(kg·d)-1氟西汀及12.8 g·(kg·d)-1百合逍遥散,空白组及模型组予等量生理盐水,各组液体量均按20 m L·(kg·d)-1计算。共给药28 d。实验过程中观察大鼠体质量变化及行为学改变,并于实验后测定大鼠海马5-HT、NE水平。结果:与模型组比较,给药2周后,3个给药组大鼠体质量明显增长(P<0.01),中西结合组大鼠水平和垂直运动明显增多(P<0.05),总液体消耗明显增加(P<0.01),糖水偏爱率明显上升(P<0.05);给药4周后,同上,并且体质量变化:中西结合组>中药组>西药组,水平运动增加:中西结合组>中药组>西药组,中西结合组大鼠糖水偏爱百分比显著高于另两组(P<0.05);3个给药组大鼠与模型组的5-HT及NE水平具有统计学差异(P<0.01),并且3个给药组海马5-HT及NE水平的趋势均为:中西结合组﹥西药组﹥中药组。结论:百合逍遥散能上调抑郁模型大鼠海马5-HT及NE水平,与氟西汀联用可增强疗效,这可能是百合逍遥散抗抑郁作用的机制之一。 Objective:Explore the effects of Baihe Xiaoyao San on behavior and hippocampus 5-HT/NE of depression rats and analyze whether exist synergism to combine with Fluoxetine. Methods :60 SD female rats were divided into 5 groups ( blank group/model group/ Baihe Xiaoyao San group/Fluoxetine group/ Baihe Xiaoyao San and Fluoxetine group ). All rats were made into depression condition with CUMS 28 days except blank group rats. From the 29th day, each group was given intragastric administration one time every day with different liquid : Fluoxetine group was given a gavage of Fluoxetine liquid 1.8 mg/( kg · d)^ -1 , bai he xiao yao san group was given with bai he xiao yao san crude drug 12.8 g/( kg · d)^ -1 ,Baihe Xiaoyao San and Fluoxetine group was given Baihe Xiaoyao San crude drug 12.8 g/( kg · d)^ -1 and Fluoxetine liquid 1.8 mg/( kg· d)^ -1 , blank group and model group were administered with equal volume nor- mal saline. The liquid volume of above 5 groups were calculated on 20 mL/( kg · d)^ -1. We watched body mass, behavior of rats during overall experiment and examined contents of hippocampus 5-HT/NE. Results : Compared with model group, body mass, consumption of saceharose and open-field movement of Baibe Xiaoyao San groups rats increased remarkably,and the effects of Baihe Xiaoyao San and Fluoxetine group were better. Conclusion:Baihe Xiaoyao San can up-regulate the hippocampus 5-HT and NE of depression rats, which is one possible mechanism of antidepressant effects, and combination with Fluoxetine can enhance curative effect.
