摘要
从青藏高原农田采集的13份土样中筛选到10株产淀粉酶活力较高菌株,其中菌株ZF-3产酶活性最高,该菌株采自定日县海拔4400 m左右处.通过菌落形态、生理生化特性和16S r RNA序列比对,鉴定该菌株为萎缩芽孢杆菌(Bacillus atrophaeus).对其粗酶液酶学性质研究表明,酶反应最适温度为60℃,最适作用p H值6.0,Ca2+、Ba2+、Zn2+、Mn2+、Fe2+等多种金属离子对酶活有明显的激活作用,Mg2+对酶活有抑制作用.
The amylase-producing strains were isolated from the soil samples collected from Qinghai-Tibetan Plateau, and among them, strain ZF-3 which was collected from Dingri County 4400 meters above sea level showed the highest amylase activity. Strain ZF-3 was identified as Bacillus atrophaeus by morphological and physiological characteristics as well as by analysis of the gene encoding the 16S rRNA. The optimal temperature and pH for the amylase activity of Bacillus atrophaeus strain ZF-3 were 60 ℃ and pH 6.0, respectively. The enzyme activity was stimulated by metal ions of Ca^2+, Ba^2+, Zn^2+, Mn^2+and Fe^2+, but was inhibited by Mg^2+.
