多西紫杉醇对宫颈癌细胞Wnt传导通路的影响

摘要 目的研究多西紫杉醇对宫颈癌Caski细胞Wnt传导通路的影响,以探讨多西紫杉醇在治疗宫颈癌中的作用机制。方法 MTT比色法检测不同浓度多西紫杉醇对人宫颈癌Caski细胞株增殖的抑制作用,计算抑制率及IC50;采用RT-PCR法和免疫组织化学法检测多西紫杉醇作用后细胞c-myc基因、β-catenin蛋白表达水平的变化。结果 0.01、0.10、1.00、10.00μg/ml多西紫杉醇作用于Caski细胞24、48、72h,其抑制强度与药物浓度及时间呈正相关。结论多西紫杉醇对宫颈癌Caski细胞生长有明显的抑制作用,可诱导Caski细胞凋亡,凋亡作用的发生可能与多西紫杉醇诱导β-catenin蛋白和c-myc mRNA表达下降有关。 Objective Study on effect of docetaxel on Wnt/β-catenin signaling pathway of cervical cancer Caski cell,and then exploring the mechanism of docetaxel treatment of cervical cancer. Methods To detect with different density of docetaxel on the proliferation of human cervical carcinoma Caski cell by MTT colorimetric assay.Calculated inhibition ratio and IC50.Experimental group added paclitaxel.Control group were not treated incubator.β-catenin protein expression were detected by immuno-histochemistry.RT-PCR was used to detect the c-myc gene expressionl. Results Caski cellwere treated with docetaxel at various concent(0.01,0.10,1.00,10.00μg/ml)for 24、48、72h,inhibition rate was 11.5%,23.6%,60.2%,68.4% respectively.Docetaxel can induce Caski cells apoptosis.In experimental group,the positive rate was 32.1%.In control group,the positive rate was 69.3%.The positive rate in experimental group was significantly decreased(P〈0.05).c-myc mRNA contents in experimental group decreased significantly compared with the control group(P〈0.05). Conclusion The effect of docetaxel inhibiting Caski cell is exact.Docetaxel can induce Caski cells apoptosis.The occurrence of apoptosis may be related to decrease of B-catenin protein and c-myc mRNA expression,induced by docetaxel.
