摘要
目的观察解郁开心丹对抑郁模型大鼠脑内去甲肾上腺素(NA)、多巴胺(DA)和5-羟色胺(5-HT)含量的影响。方法 80只SD大鼠自由饮食,自然光线下适应性喂养1周后,按照Open-Field评分方法,将得分≤20分、≥130分的大鼠剔除,剩余的大鼠随机分为6组:正常对照组、模型组、阳性药物对照组、中药高剂量组、中药中剂量组、中药低剂量组。后5组通过禁水24 h、禁食24 h、4℃冰水游泳5 min、2 h行为限制、夹尾1min、潮湿环境、昼夜颠倒,7种应激刺激每天随机安排一种,各种刺激不连续出现,连续刺激3周造模。造模后灌胃给药,阳性药物对照组给予盐酸氟西汀混悬液10 mg/d;解郁开心丹高、中、低剂量组分别给予解郁开心丹2.0,1.0,0.5 g/d(给药体积为每100 g体质量给予1.0 m L)。模型组给予等量生理盐水,持续28 d。用酶联免疫法检测大鼠脑内NA,DA,5-HT的含量。结果与模型组比较,中药高剂量组、中药中剂量组大鼠脑内NA,DA,5-HT含量明显增高且P<0.05;中药高剂量组、中药中剂量组与阳性药物对照组比较无明显差异且P>0.05。结论解郁开心丹具有与西药抗抑郁药物氟西汀同样的抗抑郁作用,且抗抑郁作用机制可能与调节大鼠脑内单胺类神经递质的平衡有关。
Objective Observe Jie Yu Kai Xin Dan within the depression model rats on norepinephrine (NA), dopamine (DA) and serotonin (5-HT) content.Methods 80 SD rats in free diet,natural light,and adaptive feeding for one week. Make the behavioral score by Open-Field scoring method. Exclude the rats who's score is ≥ 120 or ≤ 30, the last rats are randomly divided into six groups, normal control group, model group, Fluoxetine group, Chinese medicine high dose group, Chinese medicine middle dose gToup, and Chinese medicine low dose group, the last 5 groups was treated by water depriva- tion for 24 h, fasted for 24 h, 4 ~C ice water swimming 5 min, 2 h behavior restrictions, tail clamp 1 min, humid environ- ment, day and night reversed, and the rat was in different stress factors every day which is randomly decided. After 3 weeks' continuous stimulation, the depression model was made, normal control group were not treated. Intragastric administration was given after the completion of the modeling, positive control group was given fluoxetine hydrochloride suspension 10 rag/d, positive control drug's high, middle and low dose group were given Jie Yu Kai Xin Dan 2.0 g/d, 1.0 g/d, 0.5 g/d (Given 1.0 mL dose volume per 100 g body weight). Model control group were given normal saline, for 28 day. Enzyme league immune method to detect the rats NA, DA and 5-HT content. Results Conclusion Compared with model group, Chinese medicine dose group of rats in high dose group,the traditional Chinese medicine (TCM) in NA, DA and 5-HT content increased obviously and P 〈 0.05 ; Traditional Chinese medicine (TCM) in high dose group, Chinese medicine dose group compared with positive drug control group no significant difference ( P 〉 0.05). Conclusion Jie Yu Kai Xin Dan with western medicine antidepressants fluoxetine same antidepressant effect, and the antidepressant mechanism may be related to regulating single amine neurotransmitter balance in rats.
