羌活鱼不同提取物血清对骨髓间充质干细胞影响的实验研究 被引量:1

Study on Effects of Different Extraction Methods of Alpine Salamander Different Extracts of Serum on Bone Marrow Mesenchymal Stem Cells

摘要 目的:分析比较羌活鱼研粉物、水提物、酸提物及总黄酮不同提取方法对骨髓间充质干细胞(BMSCs)增殖与成骨分化的影响。方法:制备羌活鱼研粉物、水提物、酸提物,提取总黄酮,分别采用高、中、低剂量灌服大鼠制备羌活鱼研粉物、水提物、酸提物、羌活鱼总黄酮含药血清,按"人与动物体表面积等效剂量"计算所得的等效剂量,灌服后制备含药血清,以灌服等体积生理盐水制备对照血清;采用贴壁筛选法培养大鼠BMSCs,通过MTT法检测细胞增殖;与干预后的不同时间点分别检测细胞碱性磷酸酶活性、细胞RNA表达,并比较各组间差异。结果:羌活鱼研粉物、水提物及总黄酮含药血清具有刺激BMSCs增殖及促进其成骨性分化的能力,其中尤以总黄酮组最为明显。主要表现在细胞增殖、碱性磷酸酶活性、细胞RNA表达显著高于空白对照组(P<0.05)。结论:羌活鱼总黄酮能促进BMSCs增值与成骨分化。 Objective:To investigate the effects of alpine salamander medicated serum with different extraction methods of powdered material,aqueous extract,acid extract and total falconoid on proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.Methods:Prepare alpine salamander powdered material,aqueous extract,acid extract and total falconoid and they were given to rats by gavage according to high,moderate and low doses to get alpine salamander medicated serum of powdered material,aqueous extract and acid extract.Alpine salamander total falconoid was given with equivalent dose got by counting as human and animal body surface area equivalent dose and obtain total falconoid medicated serum.An equal volume of saline was given to get control serum.Culture rat bone marrow mesenchymal stem cells with method of adherent screening and the cell proliferation was detected by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide.Detect alkaline phosphatase activity of the cells and the expression of cells,RNA and compare the differences between groups.Results:Alpine salamander medicated serum of powdered material,aqueous extract,acid extract and total falconoid have the ability of stimulating cells proliferation acti vity and promoting cells to osteogenic differentiate and the total falconoid group was the best,which mainly reflected in aspects of cell proliferation,alkaline phosphatase activity of the cells and the expression of cells,RNA are all significantly higher than control group(P〈0.05).Conclusion:Alpine salamander total flavonoid can promote value-added activity and osteogenic differentiation of BMSCs.
