摘要
目的:对不同生长期粤北产广山药中尿囊素含量进行研究,确定粤北产广山药的最佳采收期。方法:采用高效液相色谱法。色谱柱为Diamonsil C18,流动相为甲醇-水(5∶95,V/V),流速为1.0 ml/min,检测波长为224 nm,柱温为30℃,进样量为20μl。分别测定不同生长期粤北产广山药中尿囊素的含量。结果:尿囊素进样量在0.218 2-1.309 2μg范围内与峰面积呈良好线性关系(r=0.999 6,n=6);精密度、稳定性、重复性试验的RSD≤1.1%;平均加样回收率为102.54%(RSD=1.43%,n=6)。粤北产广山药中尿囊素含量在生长期第1-8个月内逐渐升高,并于第8-10个月时达到峰值。结论:该方法操作简单、可靠、重复性好,可为粤北产广山药质量控制提供科学依据;以尿囊素为指标,粤北产广山药的最佳采收期为生长到第8-10个月之间。
OBJECTIVE:To study the allantoin content from wide yam produced in different growing periods produced in North Guangdong and determine the optimal harvest time. METHODS:HPLC method was adopted. The column was Diamonsil C18 with the mobile phase of methyl alcohol-water(5 ∶ 95,V/V)at the flow rate of 1.0 ml/min. The detection wavelength was 224 nm,and column temperature was 30 ℃. The sample size was 20 μl. The allantoin content from wide yam produced in different growing periods produced in North Guangdong was determined. RESULTS:There was a good linear relationship between the amount of allantoin and the peak area in the range of 0.218 2-1.309 2 μg(r=0.999 6,n=6). RSDs of precision,stability and repeatabillity tests were≤1.1%. The average recovery was 102.54%(RSD=1.43%,n=6). The allantoin content was gradually increased from the 1st month to the 8th month and it was reached the peak value during the 8th month and 10 th month. CONCLUSIONS:The method is simple,reliable,reproducible and can provide scientific reference for the quality control of wide yam produced in North Guangdong. With the index of allantoin,the optimal harvest time of wide yam produced in North Guangdong is 8th to 10 th month.
