摘要
目的探讨Eph A2和Ephrin A1在宫颈鳞状细胞癌变过程中的表达及其与临床病理因素间的关系。方法应用免疫组织化学方法,检测Eph A2和Ephrin A1在15例宫颈慢性炎症、35例宫颈上皮内瘤变(CIN)和70例宫颈鳞癌组织中的表达,并采用CD 34抗体标记微血管内皮细胞,计算微血管密度(MVD)。分析Eph A 2和Ephrin A1的表达与MVD之间的相关性及其与宫颈鳞癌临床病理特征的关系。结果 70例宫颈鳞癌组织中,Eph A2和Ephrin A1阳性率分别高于CIN组和宫颈慢性炎症组织,差异均有统计学意义(P<0.05)。Eph A2与Ephrin A1的表达与淋巴结转移和侵润深度有关(P<0.05);Eph A2与Ephrin A1的表达呈正相关(r=0.306,P<0.05)。Eph A2和Ephrin A1与年龄、FIGO分期及病理分级之间差异无统计学意义(P>0.05)。宫颈鳞癌的MVD与年龄无关,而与肿瘤分化程度、FI-GO分期、淋巴结转移及局部侵润深度有关(P<0.05)。经Spearman等级相关分析,Eph A2与Ephrin A1的表达与MVD呈显著正相关(P<0.01)。结论 Eph A2和Ephrin A1表达均与宫颈鳞癌的发生、发展密切相关,联合检测有助于宫颈鳞癌早期诊断和预后评价,可以为今后宫颈鳞癌的靶向治疗提供新的靶点。
Objective To explore Eph A2 and Ephrin A1 expression in cervical squamous cell cancer and its relationship with clinical pathological factors. Methods Eph A2 and Ephrin A1 expression were tested with immunohistochemical method in 15 cases of cervical chronic inflammation, 35 cases of cervical intraepithelial neoplasia and 70 cases of cervical squamous carcinoma tissues. Microvascular endothelial cells was marked with CD 34 antibody, and the microvascular density was calculated. Analysis of the correlation between MVD and the expression of Eph A2 and Ephrin A1,and its relationship with clinical pathological characteristics of cervical squamous carcinoma was conducted. Results In70 cases of cervical squamous carcinoma tissues, the Eph A2 and Ephrin A1 positive rate was higher than that of the CIN group and the cervical chronic inflammation group. The differences were statistically significant(P〈 0.05). The expression of Eph A2 and Ephrin A1 was associated with lymph node metastasis and invasion depth(P〈 0.05). Eph A2 and Ephrin A1 expression were positively correlated(r = 0.306, P〈 0.05). The differences of the positive expression rate of Eph A2 and Ephrin A1 in the three groups were not statistically significant(P〉0.05). The MVD of cervical squamous cell carcinomas had no association with age, but with tumor differentiation degree, FIGO staging, lymph node metastasis and the depth of local invasion(P〈 0.05). Spearman rank correlation analysis showed that there was a significant positive correlation between the expression of Eph A2 and Ephrin A1 and MVD(P〈 0.01). Conclusion The expression of Eph A2 and Ephrin A1 is closely related with the occurrence and development of cervical squamous carcinoma. The joint detection can help early diagnosis of cervical squamous carcinoma and help in prognosis evaluation. It can provide new targets for future targeted therapy of cervical squamous carcinoma.
