摘要
通过研究不同剂量甘草与白术配伍对DFMO模型小肠上皮细胞(IEC-6)增殖及p53,p21 mRNA和蛋白表达的影响,探讨甘草与白术配伍对细胞增殖影响的分子基础。采用流式细胞术分别检测药物对IEC-6细胞分裂速度及细胞周期的影响,qRT-PCR检测药物对IEC-6增殖相关基因p53,p21 mRNA的影响,Western blot检测药物对IEC-6细胞p5 3,p2 1蛋白表达的影响。白术颗粒可上调DFMO模型IEC-6细胞内p5 3,p2 1 mRNA和蛋白的表达水平;而甘草颗粒与白术颗粒不同比例配伍均能显著下调单独白术颗粒作用对DFMO模型IEC-6细胞p53,p21 mRNA和蛋白表达的影响,促进IEC-6细胞的增殖。对于DFMO模型IEC-6细胞,配伍用药甘草能调和白术对小肠上皮细胞的影响。
To study the effect of the combined administration of different doses of Glycyrrhizae Radix et Rhizoma and Atractylodis Macrocephalae Rhizoma on the proliferation of DFMO-treated intestinal epithelial cells (IEC-6) and p53, p21 mRNA and protein ex- pressions, in order to define the molecular basis for the effect of the combined administration of different doses of Glycyrrhizae Radix et Rhizoma and Atractylodis Maerocephalae Rhizoma on the cell proliferation. The effect of the drugs on the cell division rate and cell cy- cle of IEC-6 cells was detected by FCM. Quantitative Real-time PCR ( qRT- PCR) was used to analyze the effect of the drugs on mR- NA of p21 and p53 related to IEC-6 proliferation. Western blot was used to analyze the effect of the drugs on p21 and p53 protein ex- pressions of IEC-6 cells. Atractylodis Macrocephalae Rhizoma could increase p53, p21 mRNA and proteins expression in DFMO-treated IEC-6 cells. The combined administration of different ratios of Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix et Rhizoma could significantly down-regulate Atractylodis Macrocephalae Rhizoma's effect on p53, p21 mRNA and proteins expression in DFMO- treated IEC-6 cells and promote the proliferation of IEC-6 cells. The combined administration of Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix et Rhizoma could down-regulate Atraetylodis Macrocephalae Rhizoma's effect on DFMO-treated intestinal epi- thelial cells ( IEC-6 ).
