摘要
目的:研究唇香草挥发油(VOZ)的舒张血管作用及其作用机制。方法:采用BL-420生物机能实验系统,描记VOZ对二唑[4,3-a]喹喔啉-1-酮(OQD)、四乙基氯化铵(TEA)、格列苯脲(Gliben)和氯化钡(BaCl2)预处理内皮去除组和内皮完整组血管张力变化及无钙高钾液中钙离子收缩曲线。结果:VOZ对去氧肾上腺素(PE)预收缩的血管环具有浓度依赖的舒张作用,内皮去除组和内皮完整组血管舒张差异无统计学意义(P>0.05),半数最大效应浓度(EC50)为219.66 mg·L-1。ODQ、TEA、Gliben和BaCl2分别预处理不能抑制VOZ的血管舒张效应(P>0.05)。VOZ对无钙高钾液中钙离子收缩曲线均有显著的抑制作用(P<0.05),其最大浓度抑制率为44.4%。但VOZ不能抑制无钙液中PE引起的内钙释放(P>0.05)。结论:VOZ具有非内皮和浓度依赖的舒张血管作用,其机制可能与抑制外钙内流干扰胞质内钙离子平衡有关。
OBJECTIVE To investigate the vasodilatative effects of volatile oil from Ziziphora clinopodioides Lam（VOZ）on isolated rat thoracic aorta.METHODS Rat thoracic artery rings were pre-contracted with1μmol·L^-1 phenylephrine（PE）.Involvement of endothelium-dependent mechanisms were investigated by mechanically removing endothelium.K＋channels were investigated by pretreatment of artery rings with various K＋ channel inhibitors,and Ca2＋channels were investigated in artery rings incubated with Ca^2＋-free buffer and primed with PE/K＋prior to adding CaCl2 to elicit contraction.RESULTS VOZ produced concentration-dependent relaxation of artery rings at the half maximal effective concentration（EC50）of 219.66 mg·L^-1.Mechanical removal of endothelium or pretreatment with ODQ,TEA,Gliben and BaCl2,all of them had no effects on VOZ-induced response（P〉0.05 for all）.VOZ had no blocking effects on PE induced intracellular Ca2＋influx（P〉0.05）However,pre-incubation of artery rings with VOZ for 10 min produced concentration dependent inhibition on CaCl2-induced vasoconstriction by VOZ,and its maximal concentration inhibition rate was 44.40%.CONCLUSION These findings suggest the vasodilatative effects of VOZ on rat thoracic artery is independent of endothelium-derived mediators,whereas,inhibition of Ca2＋influx in vascular smooth muscle cells is important.VOZ may be a useful antihypertensive agent for some patients with hypertensive diseases.
