摘要
目的初步探讨三草黄芪汤对高血压大鼠的降压作用效果及机制。方法 50只高血压大鼠,随机分为5组即空白对照组、依那普利组、三草黄芪汤高、中、低剂量组,每组10只。灌胃给药每日1次,空白对照组注射等体积生理盐水。5周后,取血,测定血清一氧化氮(NO)及超氧化物歧化酶(SOD)含量,同时取胸主动脉组织测定内皮一氧化氮合酶(e NOS)表达。结果与空白对照组相比,三草黄芪汤治疗一周时只有高剂量可以显著降低血压,在治疗两周以后,三个剂量均可显著降低血压(P<0.05)。与空白对照组相比,三草黄芪汤可显著提高NO及SOD含量(P<0.05)。e NOS在胸主动脉表达呈逐渐增强态势。结论三草黄芪汤可显著升高高血压大鼠血清NO和SOD水平,使e NOS蛋白表达增强,判断其调节血压机制是影响胸主动脉e NOS蛋白表达,从而增加NO的含量,使血管扩张达而达到降压的效果。
Objective To investigate the antihypertensive effect of three grass astragalus root soup on hypertensive rats. Methods 50 hypertensive rats were randomly divided into five groups: control, enalapril and three grass astragalus root soup (high, middle and low dose) group. Each group has 10 cases. All rats were given intragastric administration, once a day, using normal saline in control group. Five weeks later, the expression of NO, SOD and eNOS were measured. Results Compared with control group, three grass astragalus root soup can significantly reduce the blood pressure and increase the expression of NO, SOD and eNOS (P〈0.05). Conclusion Three grass astragalus root soup can significantly increase the expression of NO, SOD and eNOS, suggesting that three grass astragalus root soup can regulate blood pressure via increasing the expression of NO by affecting the expression of eNOS..
