摘要
目的:观察丹参注射液含药血清对离体培养兔脊柱运动节段椎间盘活性及功能的影响。方法:将14只新西兰白兔随机分为空白组与含药血清组,各7只,处死后在无菌条件下取出脊柱运动节段各25个,其中空白组含有10%无药血清,含药血清组含有10%丹参注射液含药血清,两组于培养前及培养后第3、7、14、21天,各取5个椎间盘分别进行HE染色、Ⅱ型胶原免疫组化、蛋白多糖含量测定。结果:培养14d空白组椎间盘组织形态出现部分破坏,含药血清组组织形态维持良好,21d两组组织形态出现破坏;培养14d两组Ⅱ型胶原免疫组化染色强度无显著差异,21d时均出现显著下降(P<0.05),且空白组较含药血清组下降明显(P<0.05);培养7d两组蛋白多糖PAS/AB染色强度无降低,14d两组强度均有所减弱,21d进一步减弱,且空白组改变更为明显。结论:空白组14d以后椎间盘逐渐出现退变,21d时退变明显;含药血清组培养14d仍可维持椎间盘的活性与功能,21d时也出现退变,但退变程度不如空白组显著,提示含有丹参注射液的药物血清可以一定程度上减缓椎间盘退变。
Objective: To observe the effects of Salvia Injection containing serum on the intervertebral discs of in vitro culture rabbit spinal motion segment. Methods: Twenty-five spinal motion segments were harvested respectively under aseptic conditions from 14 New Zealand white rabbits which were randomly divided into blank control group and Salvia Injection containing serum group with 7. The DMEM of blank control group contained 10% blank serum, and DMEM of Salvia Injection containing serum group contained 10% Salvia Injection serum, then 5 discs of the two groups were observed respectively by HE staining, immunohistochemistry of collagen type Ⅱ, proteoglycan content before culture and on 3th, 7th, 14th, 21th days after culture.Results: HE showed after culture of 14 days there were partial destruction on the intervertebral disc tissue structure of blank control group, and the structure remained intact of Salvia Injection containing serum group, however on 21 days there was severely degenerated of two groups; The intensity value of type Ⅱ collagen immunohistochemical staining in the nucleus pulposus were not changed significantly within 14 days, but the staining intensity became shallower on 21 days of two groups (P〈0.05), and the change of blank control group was more obvious (P〈0.05); PAS/AB staining of proteoglycan of nucleus pulposus showed that there were not decrease of tinting strength of two groups on 7 days, but the strength weakened slightly of two groups on 14 days, on 21 days the tinting strength became weaker, and the change of blank control group Was more significant. Conclusion: After cultured 14 days, there is gradually appeared degeneration of the blank control group, and which has significant degeneration on 21 days; Meanwhile it is not obviously degenerated of the discs of Salvia Injection containing serum group cultured within 14 days, although there is degeneration on 21 days, the degree of degeneration of blank control group is more significant. These findings suggests that the serum contained Salvia Injection could in the certain degree postpone the degeneration of the intervertebrai disc.
