采用小鼠模型探讨行囊胚活检后胚胎的冷冻时机

摘要 【目的】探讨行囊胚活检后,活检胚胎最佳的冷冻时机。【方法】258枚小鼠优质囊胚进行囊胚活检,分别在活检后0 h(n=43),0.5 h(n=39),1 h(n=50),2 h(n=49)进行玻璃化冷冻,仅行活检的新鲜囊胚(n=48)以及冷冻的未活检囊胚(n=29)分别作为对照组,观察各组的复苏率、重新扩张率和孵出率,并计数囊胚细胞总数、凋亡细胞数、囊胚DNA完整率和活检部位凋亡细胞数。【结果】各组的存活率、重新扩张率和孵出率无差别;平均囊胚细胞总数、凋亡细胞数、囊胚DNA完整率和活检部位凋亡细胞数差别无统计学意义。【结论】小鼠囊胚活检后不同时间点冷冻效果未观察到差异,可根据工作需要灵活安排活检囊胚的冷冻时间。 【Objective】 To try to find an optimal time to vitrify biopsied blastocyst using mouse model. 【Methods】 258 mouse blastocysts were divided into four treatment groups and two control groups(biopsy control group(n = 48) and vitrification control group(n = 29). Biopsied blastocysts were cryopreserved after different time of in vitro culture [0 h(n = 43), 0.5 h(n = 39), 1 h(n= 50), 2 h(n = 49)]. Blastocyst cryo-survival rate, blastocele re-expansion rate and blastocyst hatching rate were compared among groups. The mean total blastocyst cell number, apoptotic cell number, DNA integrity index and the apoptotic cells at biopsied site were counted. 【Results】 The cryo-survival rate, blastocele re-expansion rate and hatching rate were not statistically different among groups; The mean total blastocyst cell number, apoptotic cell number, DNA integrity index and the apoptotic cells at biopsied site were not statistically different among groups too. 【Conclusion】 In vitro culture before vitrification did not show any beneficial effect for the cryopreservation of biopsied blastocysts. Cryopreservation of biopsied blastocysts could be carried out flexibly in IVF lab after biopsy manipulation.
