2011—2012年中国甲型H3N2亚型流感病毒对神经氨酸酶抑制剂药物的敏感性 被引量:6

Susceptibility of human influenza A (H3N2) viruses to neuraminidase inhibitors isolated during 2011-2012 in China

摘要 目的分析2011—2012年中国甲型H3N2亚型流感病毒对神经氨酸酶抑制剂药物(NAI)的敏感性。方法所有待检流感毒株来自中国流感监测网络,该网络覆盖中国31个省份的408家网络实验室和554家哨点医院。本研究挑选了2011年1月1日至2012年12月31日采样分离的毒株,共1903株,其中经国家流感中心复核鉴定为甲型H3N2亚型流感病毒的毒株共721株,采用化学发光法检测其对奥司他韦和扎那米韦的敏感性。药物敏感参考毒株为A/Washington/01/2007(119E),奥司他韦耐药参考毒株为A/Texas/12/2007(E119V)。采用t检验比较不同年份毒株对NAI敏感性的差异。结果A/Washington/01/2007对奥司他韦和扎那米韦的半数抑制浓度(IC50)值分别为(0.10±0.02)、(0.30±0.05)nmol/L,A/Texas/12/2007对奥司他韦和扎那米韦的IC50值分别为(4.27±1.60)、(0.20±0.03)nmol/L。721株甲型H3N2亚型流感病毒中,2011年132株,2012年589株。2011和2012年的甲型H3N2亚型毒株对奥司他韦的IC50值范围分别为0.04~0.62、0.02~0.95nmol/L,均在A/Washington/01/2007的IC50值10倍(1.00nmol/L)以内。2011和2012年的甲型H3N2亚型毒株对扎那米韦的IC50值范围分别为0.12~0.80、0.04~0.72nmol/L,均在A/Washington/01/2007的IC50值10倍(3.00nmol/L)以内。结论2011—2012年检测的中国甲型H3N2亚型流感毒株均对奥司他韦和扎那米韦敏感。 Objective To analyze the susceptibility of influenza A (H3N2) viruses to neuraminidase inhibitors during 2011-2012 in China's Mainland. Methods All the tested viruses were obtained from the Chinese National Influenza Surveillance Network, which covers 31 provinces in China's Mainland, including 408 network laboratories and 554 sentinel hospitals. In total 1 903 viruses were selected with isolation date from January 1,2011 to December 31, 2012 in China's Mainland, among these viruses, 721 were confirmed to be influenza A (H3N2) virus by Chinese National Influenza Center and tested for the susceptibility to oseltamivir and zanamivir using chemiluminescence-based assay. The neuraminidase inhibitor sensitive reference virus A/Washington/01/2007(119E) and oseltamivir resistant virus A/Texas/12/ 2007(El19V) were used as control in this study. The t-test was used to compare the difference of NAI susceptibility of viruses isolated from different years. Results The half maximal inhibitory concentration (IC50) of A/Washington/01/2007 for oseltamivir and zanamivir was (0.10± 0.02) and (0.30 :t: 0.05) nmol/L, respectively. The IC50 of Afrexas/12/2007 for oseltamivir and zanamivir was (4.27 ±1.60) and (0.20±0.03) nmol/L, respectively. Among the 721 influenza A (H3N2) viruses, 132 influenza A(H3N2) viruses were isolated in 2011 and 589 influenza A (H3N2) viruses were isolated in 2012. The IC50 for oseltamivir ranged from 0.04 to 0.62 nmol/L for viruses isolated in 2011 and ranged from 0.02 to 0.95 nmol/L for viruses in 2012, and the IC50 of all the viruses tested was within 10-fold IC50 (1.0 nmol/L) of the neuraminidase inhibitor sensitive reference virus A/Washington/01/2007. The IC50 of zanamivir ranged from 0.12 to 0.80 nmol/L for viruses in 2011 and ranged from 0.04 to 0.72 nmol/L for viruses in 2012, and was within 10-fold IC50 (3.0 nmol/L) of the neuraminidase inhibitor sensitive reference virus A/Washington/01/2007. Conclusion The influenza A(H3N2) viruses isolated during 2011-2012 in China's Mainland were tested to be sensitive to oseltamivir and zanamivir.
