摘要
以2个高油品系(扬36、扬3052)和一个低油品系(扬6002)发育过程中的甘蓝型油菜种子为材料,利用实时荧光定量PCR技术,比较9个参与脂肪酸合成的编码酶基因和3个相关转录因子在种子发育过程中的表达模式和表达水平差异,探讨不同含油量甘蓝型油菜油脂合成代谢的差异。结果表明:除ACCase、FAD3、LEC1外,不同高油品系之间多数基因的表达模式和表达量相近,说明这2个不同遗传来源的高油品系在油分形成过程中,其油脂合成的主要基因在表达上具有较好的一致性。但高油和低油品系之间多数基因的表达模式或表达水平则存在着明显差异,其中FatA、FAD3、L1L等基因的表达量与含油量密切相关,随种子的发育其在高油品系中表达量明显高于低油品系。
In order to investigate the differences of lipid synthesis gene expression between high and low oil content rapeseed, two high oil content lines (Yang 36, Yang 3052) and one low oil content line (Yang 6002) were analyzed by quantitative Real-time PCR. The expression patterns and levels of 9 enzyme coding genes and 3 transcription factors involved in lipid synthesis during seed development were analyzed. The results showed that most of genes had similar expression patterns and levels (except ACCase, FAD3 and LEC1) in two high oil content lines, indicating the key genes expression of lipid synthesis had good consistency between these two high oil content lines with different genetic source. However, the expression patterns and levels of most genes were different between high and low oil content rapeseed. The expression of some genes such as FatA, FAD3, L1L was closely related to oil content, and with the development of seeds, they showed higher expression levels in high oil content line than those in low oil content line, suggesting that they may play a positive role in increasing oil content.
