摘要
目的:观察癃畅颗粒对前列腺增生细胞增殖和凋亡及相关因子的影响。方法:将雄性Wistar大鼠100只,严格遵循随机分组的原则随机分为正常组、模型组、癃畅颗粒高、中、低剂量组,各20只。模型组、癃畅颗粒高、中、低剂量组给予前列腺增生模型复制。正常组:普通饲养,按1 m L/100 g体重给予生理盐水灌胃,日1次,连续30 d;模型组从去势第8天起,按1 m L/100 g体重给予生理盐水灌胃,日1次,连续30 d;癃畅颗粒低、中、高剂量组:从去势第8天起,按1 m L/100 g体重分别给予低(100 mg/m L)、中(200 mg/m L)、高浓度(300 mg/m L)灌胃,日1次,连续30 d。观察前列腺组织前列腺湿重和前列腺指数、前列腺细胞增殖指数和凋亡指数、VEGF、IGF-1 m RNA、Bcl-2和Bax表达变化。结果:与正常组比较,模型组和癃畅颗粒高、中、低剂量组前列腺湿重和前列腺指数明显升高,有统计学意义(P<0.05);与模型组比较,癃畅颗粒中、高剂量组大鼠的前列腺湿重和前列腺指数显著下降,有统计学意义(P<0.05)。与正常组比较,模型组大鼠和癃畅颗粒高、中、低剂量组前列腺增生细胞的凋亡指数明显升高,有统计学意义(P<0.05),而增殖指数没有显著性差异(P>0.05);与模型组比较,癃畅颗粒高、中、低剂量组大鼠前列腺增生细胞的凋亡指数明显升高,有统计学意义(P<0.05),而增殖指数没有显著性差异(P>0.05),与正常组大鼠前列腺组织中上皮细胞层Bcl-2蛋白阳性表达比较,模型组和癃畅颗粒高、中、低剂量组,有统计学意义(P<0.05),模型组表达最强,高剂量组最弱,与正常组接近,模型组和癃畅颗粒高、中、低剂量组Bcl-2蛋白阳性表达比较,有统计学意义(P<0.05)。与正常组上皮细胞层Bax蛋白阳性表达比较,模型组和癃畅颗粒低剂量组,有统计学意义(P<0.05),与模型组上皮细胞层Bax蛋白阳性表达比较,癃畅颗粒高、中剂量组,有统计学意义(P<0.05)。与正常组VEGF表达相比,模型组和癃畅颗粒高、中、低剂量组升高最为显著,有统计学意义(P<0.05);与模型组相比,癃畅颗粒高、中、低剂量VEGF表达降低,有统计学意义(P<0.05)。与正常组IGF-1 m RNA表达相比,模型组和癃畅颗粒高、中、低剂量组升高最为显著,有统计学意义(P<0.05);与模型组相比,癃畅颗粒高、中剂量IGF-1 m RNA表达降低,有统计学意义(P<0.05)。结论:癃畅颗粒可以有效下降前列腺湿重和前列腺指数,降低VEGF、IGF-1m RNA、Bcl-2和提高Bax表达,促进前列腺细胞凋亡。
Objective : To observe the Longehang granule on hyperplasia of prostate and related factors of cell proliferation and apoptosis. Methods : 100 male Wistar rats, strictly follow the principle of random grouping were randomly divided into normal group, model group, Longchang particles high, medium and low dose group, each 20. Model group, Longchang particles high, medium and low dose group was given the prostate hyperplasia model replication. Normal group: normal feeding, press 1 mL/lO0 g weight for saline lavage, day 1 times, 30 d; From castrated model group 8, press 1 mL/100 g weight for saline lavage, day 1 times, 30 d ; Longehang particles of low, medium and high close groups : since castration day 8, according to the 1 mL/100 g weight were given low ( 100 mg/mL ), medium ( 200 mg/mL ), high concentration ( 300 mg/mL ) to fill the stomach, day 1 times, 30 d. Observe prostate tissue of prostate tissue wet weight of prostate and prostate index and prostate cell proliferation and apoptosis index, VEGF mRNA, IGF-1, the Bel-2 and Bax expression changes. Results: Compared with normal group, model group and Longehang particles high, medium and low dose group of wet weight of prostate and prostate index increased significantly, with statistical significance (P〈0.05); Compared with model group, Longchang particles, high dose group of rats in a significant reduction in the wet weight of prostate and prostate index, was statistically significant ( P〈0.05 ). Compared with normal group, model group rats and Longchang particles high, medium and low dose group of hyperplasia of prostate cells apoptosis index increased significantly (P〈0.05), while there is no significant difference between the proliferation index ( P〉0.05 ); Compared with model group, and Longchang particles high, medium and low dose group of rat prostate cells apoptosis index increased significantly, with statistical significance (P〈0.05), while there is no significant difference between the proliferation index ( P〉0.05 ), and normal group of epithelial cells in the prostate tissue of the layer of the Bcl-2 protein positive expression, model group and Longehang particles high, medium and low dose group, with statistical significance (P〈0.05), the strongest expression of model group, high dose group of the weakest, close to the normal group, model group and Longchang particles high, medium and low dose group of the Bcl-2 protein positive expression, with statistical significance ( P〈0.05 ). Group and normal epithelial layer Bax protein positive expression, model group and low dose group, Longehang grain was statistically significant ( P〈0.05 ), and the model group epithelial layer Bax protein positive expression, Longchang particles of high, middle dose group, with statistical significance ( P〈O.05 ). VEGF expression compared with normal group, model group and Longchang particles rise high, medium and low dose group of the most significant, was statistically significant (P〈0.05); Compared with model group, Longehang particles high, medium and low doses of VEGF expression is reduced, with statistical significance ( P〈0.05 ). Compared with normal group of IGF-1 mRNA expression, model group and Longchang particles rise high, medium and low dose group of the most significant, was statistically significant (P〈0.05); Compared with model group, Longchang particles of high, middle dose of IGF 1 mRNA expression decreased, with statistical significance ( P〈0.05 ). Conclusion : Longchang particles can effectively decline the wet weight of prostate and prostate index, reduce VEGF mRNA, IGF-1, the Bcl-2 and increase the expression of Bax, promote apoptosis of prostate.
