摘要
目的通过观察房颤患者miR-208及炎性因子IL-18水平变化,分析miR-208与IL-18及房颤相关性,进一步探讨房颤的发生机制。方法选择99例房颤患者为房颤组(32例阵发性房颤、31例持续性房颤、36例永久性房颤),29例健康志愿者为对照组,运用实时荧光定量聚合酶链式反应法检测2组血浆miR-208的表达水平、酶联免疫法检测IL-18水平,分析miR-208与心房内径大小及IL-18水平相关性及临床意义。结果与对照组比较,房颤组血浆miR-208、IL-18的表达水平明显升高,差异有统计学意义(P<0.05);房颤组中,与阵发性房颤比较,持续性房颤及永久性房颤组血浆miR-208、IL-18的表达水平明显升高,差异有统计学意义(P<0.05),且随房颤持续时间的延长表达增加。血清miR-208水平与心房内径、IL-18呈正相关(r=0.378、0.365,P<0.05),偏相关分析校正后它们之间仍然呈正相关(r=0.364、0.353,P<0.05)。结论房颤患者miR-208与心房内径、IL-18关系密切,相互关联,通过探讨房颤患者miR-208、IL-18水平及关系机制,为房颤的诊疗提供新思路。
Objective To observe the changes of levels of miR-208 and interleukin-18( IL-18) in patients with atrial fibrillation( AF),and to analyze the correlation between miR-208 and IL-18 so as to explore the pathogenesis of AF. Methods Ninety-nine patients with AF were served as AF group including 32 cases of paroxysmal AF,31 cases of persistent AF and36 cases of permanent AF,and 29 healthy volunteers were served as control group. The expression levels of plasma miR-208 were detected by fluorescent quantitation Real-time polymerase chain reaction( qRT- PCR) and the levels of IL-18 were detected by ELISA for both groups,moreover,the clinical significance and correlation between miR-208 and internal diameter of cardiac atrium as well as IL-18 levels were analyzed. Results As compared with those in control group,the expression levels of plasma miR-208,IL-18 in AF group were significantly increased( P 〈0. 05). In AF group,as compared with those in paroxysmal AF,the expression levels of plasma miR-208,IL-18 in persistent AF and permanent AF were significantly increased( P 〈0. 05),moreover,which were increased more obviously with the duration of AF. The expression levels of serum miR-208 were positively correlated to internal diameter of cardiac atrium and IL-18 levels( r = 0. 378,0. 365,respectively,P 〈0. 05),furthermore,after revising partial correlation analysis,the positive correlation was still remained( r = 0. 364,0. 353,respectively,P 〈0. 05). Conclusion The expression levels of miR-208 and IL-18 are closely correlated to internal diameter of cardiac atrium and IL-18 levels,which provides new ideas for the treatment of AF.
