摘要
以细菌菌落计数方法,研究大肠埃希菌、金黄色葡萄球菌、鼠伤寒沙门氏菌在不同2,3,5-三苯基氯化四氮唑(TTC)浓度的平板计数琼脂上菌落形态和数量的变化情况。结果表明,3种菌在TTC终浓度≥0.0008%时,菌落颜色比较明显,逐渐加深;大肠埃希菌在TTC终浓度≤0.01%的平板计数琼脂上的菌落数量和对照组无显著差异,TTC终浓度≥0.02%菌落数量明显减少;金黄色葡萄球菌在TTC终浓度≤0.004%的平板计数琼脂上的菌落数量和对照组无显著差异,在0.04%TTC平板计数琼脂上不能生长;鼠伤寒沙门氏菌TTC终浓度≤0.01%的平板计数琼脂上的菌落数量和对照组无显著差异,在TTC终浓度≥0.02%的平板计数琼脂上菌落数量减少。不同浓度的TTC对鼠伤寒沙门氏菌、大肠埃希菌的生长影响较小,对金黄色葡萄球菌影响较大,故在食品(或饲料)细菌总数检测中,TTC的终浓度应选在0.0008%~0.004%之间。
According to bacterial colony counting method ,we researched the changes of bacterial colony morphology and quantity of Escherichia coli ,Staphylococcus aureus and Salmonella typhimurium in different concentrations of TTC agarplate .The results showed that the color of bacterial colony of these three strains was obvious and became darkening when TTC final concentration ≥0.0008% ;In TTC final concentration ≤0.01% ,the bacterial colony number of E .coli was no significant difference from that of the control group .However ,the number of colonies were significantly reduced in TTC final concentration ≥0.02% ;In TTC final concentration ≤0.004% ,the bacterial colony number of Staphylococcus aureus was no significant difference from that of the control group;In addition ,the bacterial colony could not grow in the medium of TTC's concentration reaching to 0.04% ;When TTC final concentration ≤0.01% ,the bacterial colony number of Salmonella typhi was no significant difference from that of the control group .But when TTC final concentration ≥0.02% ,the number of colonies was reduced .Conclusions:Different TTC concentrations have less effect on the growth of Salmonella typhi and E .coli ,while they have greater impact on the growth of Staphylococcus aureus .So in the detection of total bacteria number in food(or fodder) ,TTC final concentration should be chosen between 0.0008% and 0.004% .
