摘要
目的:验证胶质瘤细胞(C6)可能的一种乳酸快速排泌机制。方法:C6按6组不同浓度(0.5×105;0.75×105;1×105;1.25×105;1.5×105;2×105)接种,24h收集上清液,检测乳酸,乳酸脱氢酶含量,以海马神经元HT22作为对照。结果:接种浓度为1×105个/L组细胞24h密度适中处于指数增生期,乳酸和乳酸脱氢酶含量高于其他组(P<0.01)(除1.25×105个/L浓度组)。C6细胞培养上清液中乳酸含量明显高于HT22细胞培养上清液(P<0.01)。结论:胶质瘤细胞可能有一种快速排泌机制,使得其代谢产生的大量乳酸在细胞外液中堆积。
Objective: To validate the quick exhaust urinary mechanism of lactic acid glioma cell.Methods:C6cells were inoculated according to different concentrations in 6 groups(0.5 × 10^5; 0.75 × 10^5; 1 × 10^5; 1.25 ×10^5; 1.5 × 10^5; 2 × 10^5).The supernatant was collected after 24 hours.The contents of lactic acid and lactate dehydrogenase were determinated with hippocampal neurons HT22 as control.Results: The number of cells is moderate and cells grow at an exponential rate in the group of 1 × 10^5/ L inoculation concentration after 24 h.Lactic acid and lactate dehydrogenase were higher than those in other groups(P 0.01)(except for 1.25 × 10^5 concentration group).Lactic acid content of C6 cell culture supernatant was significantly higher than that in HT22cell(P 0.01).Conclusion: Glioma cells may have a rapid excretion mechanism.The accumulation of large amounts of lactic acid makes the metabolized in the extracellular fluid.
