摘要
目的研究乌司他丁对致死性烫伤大鼠心肌乙酰化组蛋白H3赖氨酸9(Ac-H3K9)表达水平的影响及意义。方法 80只雄性SD大鼠按随机数字表法分为烫伤组和乌司他丁组(n=40),采用80℃水浴浸泡背部15 s、双下肢15 s、腹部8 s,两组动物均造成50%总体表面积Ⅲ度烫伤大鼠模型。烫伤后即刻静脉分别注射生理盐水和乌司他丁40 000 U/kg。各组分别于烫伤后4 h取大鼠腹主动脉血,测定血浆肌酸激酶同工酶(CK-MB)水平;然后处死动物取心肌组织,用光学显微镜观察大鼠心肌组织的病理改变;用蛋白质免疫印迹法(Western blot)测定心肌细胞中Ac-H3K9的表达。结果与单纯烫伤组伤后4 h CK-MB[(1 571.8±251.4)U/L]相比,烫伤后4 h乌司他丁组为(938.3±140.7)U/L,显著降低(P<0.05);伤后4 h乌司他丁组Ac-H3K9表达水平均显著高于烫伤组(P<0.05)。结论乌司他丁能对致死性烫伤大鼠早期的心肌损害早期有保护作用,其机制可能与乌司他丁能增加心肌保护性蛋白Ac-H3K9的表达有关。
Objective To investigate the expressions ofhistone H3 on ulinastatin myocardium in rats following lethal burn injury and its relationship with the expression of protective protein. Methods Eighty male Sprague-Dawley rats were randomly divided into scald group and ulinastatin group. Rats in the two groups were subjected to 50% total body surface area third-degree burns. Immediately after scald rats were intravenously given saline in scald group or saline containing ulinastatin (40 000 U/kg) in ulinastatin group. Blood of rats in each group was withdrawn from the abdominal aorta at 4 hours after injury for measurement of plasma creatine kinase-MB (CK-MB) activities; The rats were sacrificed at the end of experiment and observed the pathological change of myocardial tissue in rats by optical microscope. Heart tissues were harvested for the measurement of acetylated histone H3 by Western blot. Results The CK-MB after 4 h injury in ulinastatin group was (938.3±140.7)U/L, significantly lower than those (t 571.8±251.4)U/L in scald group (P〈0.05). The expression of histone H3 after 4 h injury in ulinastatin group were obviously higher than those in scald group (P〈0.05). Conelusions The results indicated that ulinastatin significantly alleviate myocardial injury in early stage of bum injury. These protective effects may be due its increasing expressions of histone H3 may be related to improvement of the tolerance heart to burn injury.
