人羊膜间充质干细胞抑制肝损伤小鼠淋巴细胞增殖和干扰素-γ分泌的研究

Study on the human amniotic mesenchymal stem cells inhibiting lymphocyte proliferation and interferon-gamma secretion in mice with liver injury

摘要 为了研究人羊膜间充质干细胞(h AMSCs)进入损伤动物体内后对动物机体免疫相关指标的影响,探究人羊膜间充质干细胞免疫原性,试验采用两步酶消化人羊膜组织分离培养h AMSCs,用流式细胞技术和免疫荧光的方法鉴定细胞表面分子;将CCl4橄榄油混合液按体重比腹腔注射到小鼠体内造肝损伤模型,损伤后的小鼠尾静脉注射h AMSCs,观察小鼠30 d内外周血中淋巴细胞和血浆中γ-干扰素(IFN-γ)的变化。结果表明:试验分离得到较高纯度的h AMSCs细胞;尾静脉注射细胞的肝损伤小鼠与未注射细胞的肝损伤小鼠相比,注射细胞的小鼠体内淋巴细胞数量有所降低;损伤小鼠体内血浆中IFN-γ含量随着h AMSCs注入时间的延长也逐步降低。h AMSCs进入肝损伤小鼠体内不引起淋巴细胞发生增殖,也不诱发分泌IFN-γ,反而降低动物体内的免疫功能。说明h AMSCs具有低免疫原性,可能使其进入机体后不被免疫细胞识别,从而能够在机体内修复脏器。 To study the effect of human amniotic mesenchymal stem cells( h AMSCs) on the immune- related indicators in animals' bodies after entering the injured animals,and investigate the immunogenicity of h AMSCs,a two- step enzymatic digestion was used to digest human amniotic membrane tissue for isolation and culture of h AMSCs. The cell surface molecules were identified by flow- cell technology and immunofluorescence method. The mixture of olive oil with carbon tetrachloride was used for intraperitoneal injection into the mice to make a liver injury model while the h AMSCs were injected into the injured mice through the tail vein,and then the changes in lymphocytes and plasma interferon- γ( IFN- γ) in peripheral blood of mice were observed for 30 d,respectively. The results showed that higher purity of h AMSCs was obtained by isolation; and compared to the control group of liver damage mice,the number of lymphocytes in the injected mice from the liver damage group decreased somewhat compared with that in the liver damage group without injecting the h AMSCs; The IFN- γ contents in plasma of injured mice gradually reduced with extension of the injected time of h AMSCs; The h AMSCs entering the bodies of mice with liver injury did not cause lymphocyte proliferation in vivo,and did not induce the secretion of IFN- γ,but decreased the immune function in mice. The results indicate that the h AMSCs have low immunogenicity,which makes them not to be recognized by the immune cells after entering the bodies of mice.Thus,it is possible to repair the organs in the body.
