HPLC-ELSD法建立小柴胡汤正丁醇有效部位的指纹图谱 被引量:2

摘要 目的建立小柴胡汤正丁醇有效部位整体特征的质量评价方法。方法采用高效液相蒸发光散射检测器,Diamonsil C18(2)(250mm×4.6mm,5μm)色谱柱,以乙腈-体积分数为0.05%的甲酸水溶液为流动相进行梯度洗脱,建立小柴胡汤和各药材正丁醇有效部位指纹图谱。结果经评价软件计算相似度大于0.9,对21个共有峰进行了药材归属,并指认了其中的6个色谱峰。结论建立的小柴胡汤正丁醇有效部位的HPLC-ELSD指纹图谱准确稳定、重复性好,且能较好地判别有效部位主要色谱峰与药材化学组分间的相关性,为阐明有效部位的化学物质基础提供科学依据。 Objective To establish a quality evaluation method for the overall features of the n-butanol effec- tive fraction of Xiao Chaihu Tang. Methods The chromatographic fingerprints of the effective fraction and the relevant fractions of its herbs were configured by HPLC/ELSD analysis. The separation was performed on Diamonsil C18(2) (250 mm ×4.6 mm,5 μm). The mobile phase consisted of acetonitrile and 0.05% (φ) methanoic acid was used as gradient elution for separation with HPLC at a flow rate of 1 mL. min-1. Results The fingerprint of n-butanol fraction of Xiao Chaihu Tang was successfully established and 21 common fin- gerprint peaks were marked and attributed to the herbs. Six chromatographic peaks were identified. Conclu- sions The established HPLC-ELSD fingerprint is accurate, stable, and reproducible, which could reflect the o- verall characteristics of the n-butanol effective fraction of Xiao Chaihu Tang. The fingerprint of n-butanol fraction is helpful to state chemical material foundation.
