摘要
目的建立测定蒙药金诃子低温提取鞣质成分和含量的测定方法。方法采用红外光谱技术和磷钼钨酸-干酪素比色法测定五个不同部位提取物中鞣质的成分和含量。红外光谱技术检测部分由清华大学分析中心分析;磷钼钨酸-干酪素比色法以没食子酸为对照品,选取波长和检测时间点,并制备标准曲线进行检测。结果红外光谱技术测得五个部位提取物在单宁酸的特征吸收谱带中均有不同程度的吸收峰,而单宁酸是鞣质的主要成分,但是不同部位之间鞣质含量差异无显著性;磷钼钨酸-干酪素比色法测定波长选定为760 nm;在30 min和45 min时间点结果均较稳定,所以选择在此时间段内测定鞣质含量;标准曲线线性回归方程为Y=0.136 2X-0.086 3,R=0.996 1;检测五个不同部位提取物含量分别为68.81±2.78%、25.16±0.89%、56.83±0.90%、20.47±1.21%、32.82±1.54%,鞣质含量最少的4号部位与1号和3号部位比较差异有统计学意义,P<0.05。结论蒙药金诃子低温提取的五个不同部位,成分分析优选红外光谱技术,而鞣质含量检测优选磷钼钨酸-干酪素比色法。
Objective To establish a method for component and content determination of tannins in Mongolian medicine Terminalia chebula Ritz. low temperature extraction. Methods The component and content determination of tannins in five different positions were determined by infrared spectroscopy and tungsten molybdophosphate- casein colorimetric method. Experimental portion of infrared spectroscopy detection was completed in tsinghua university analysis center; Tungsten molybdophosphate- casein colorimetric method was used with gallic acid as the standard, and wavelength selection and detection time were decided in the experiment, then standard curve was prepared to detect the content of tannins. Results The component of tannins in five different positions were determined by infrared spectroscopy, but the content was no significant difference among five different positions; In tungsten molybdophosphate-casein colorimetric method, the wavelength selection was 760 nm, and detection results were stable at 30 min and 45 min; linear regression equation of standard curve was Y = 0.136 2X- 0.086 3,R = 0.996 1; there were tannins in five different positions, and the content of tannins in five different positions were 68.8 ± 2.78%, 25.1 ± 0.89%, 56.8 ± 0.90%, 20.4 ± 1.21%, 32.8 ± 1.54%. Conclusion The component determination of tannins in five different positions was preferably detected by infrared spectroscopy, and the content determination of tannins was by tungsten molybdophosphate- casein colorimetric method.
