摘要
对木聚糖酶嗜热真菌嗜热踝节菌F1208的产酶条件进行优化,经单因素试验及Box—Behnken响应面分析,确定该菌液体发酵培养基,其组成是:碳源为玉米芯粉,碳源粒度24—40目,碳源质量分数5%;氮源为大豆蛋白胨与酵母膏复合氮源(质量比l:3),氮源质量分数2%;辅助添加物甘油(0.2%);培养基初始pH6.4。优化后最佳产酶条件是:装液量60mL,培养温度48℃,摇床转速200r/min。在此条件下发酵培养7d,产酶量迭3026.65U,mL,是初始酶活(960.39U/mL)的3.15倍,为同类研究中较高水平,具有较好的应用前景。
The culture conditions of a thermophilic fungus F1208 with high yield of xylanase activity were optimizedby single-factor experiment and Box-Behnken design matrix of response surface methodology. The influences of medium composition and fermentation conditions in the cultivation were investigated. A high xylanase activity of 3 026.65 U/mL was obtained in 7 days, which was 3.15 fold of the initial enzyme activity (960.39 U/mL). The results indicated optimum conditions were corncob, mesh number 24-40, 5%; soya peptone and yeast cream (ratio 1:3) 2%; glycerine 0.2%; pH 6.4, and the fermentation conditions was 60 mL, 48℃ and 200 r/min.
