Simultaneous Determination of Aloe-emodin and Physcion in Wikstroemia indica by HPLC

摘要 目的:建立HPLC法同时测定了哥王中芦荟大黄素和大黄素甲醚含量的方法。方法:采用C18色谱柱(4.6 mm×250 mm,5μm),流动相A为乙腈,流动相B为体积分数为0.15%磷酸水,梯度洗脱,流速1.0 m L·min^-1,检测波长254 nm,柱温30℃。结果:芦荟大黄素和大黄素甲醚的质量浓度分别在1.584-25.34(r=0.999 2)和1.598-25.57μg·m L^-1(r=0.999 3)内与峰面积呈良好的线性关系,平均回收率分别为100.2%(RSD 1.8%)和99.7%(RSD 1.1%)。不同产地了哥王中大黄素甲醚和芦荟大黄素含量差异较大,其中广东韶关产地的芦荟大黄素含量最高,大黄素甲醚含量最低;福建福州产地的大黄素甲醚含量最高,芦荟大黄素含量最低;浙江(批号120724)了哥王未检测到芦荟大黄素,贵州安顺产地的了哥王中未检测到大黄素甲醚。结论:该方法简便、快速、准确,为了哥王药材的质量控制提供参考。 Objective: To develop an HPLC method for the simultaneous determination of aloe-emodin and physeion in Wikstroemia indica. Method: Diamonsil Clscolumn (4. 6 mm ×250 mm, 5 μm) was eluted with the mobile phases of acetonitrile-0. 10% phosphoric acid water in a gradient mode. The UV detection wavelength was 254 nm. The flow rate was 1.0 mL ·min^-1 The column temperature was set at 30 ℃ , and the sample size was 20 μL. The difference in the content of aloe-emodin and physcion in W. indica from from ten origins was compared. Result: All of aloe-emodin and physcion samples showed a good linearity within the range of 1. 584-25.34 mg· L^-1 (r = 0. 999 2), 1. 598-25.57 mg ·L^-1 (r = 0. 999 3), respectively. The average recoveries were 100. 2% and 99.7% , with RSD of 1.8% and 1.1% , respectively. There was a great difference in the content of aloe-emodin and physeion in W. indica of different origins. Specifically, W. indica from Shaoguang, Guangdong province, had the highest content of aloe-emodin but the lowest content of physcion; W. indica from Fuzhou, Fujian province, had the highest content of physeion but the lowest content of aloe-emodin. Aloe-emodin was not detected in 120724 W. indica from Zhejiang, and physcion was not detected from Anshun, Guizhou province. Conclusion: The method is simple, rapid and accurate and so can be applied for the quality control of W. indica.
