摘要
目的观察逐瘀通脉胶囊联合针刺对脑卒中大鼠皮质区病理变化的影响。方法将50只健康雄性SD大鼠随机分为5组:对照组(A)、模型组(B)、通脉组(C)、针刺组(D)及针药联合组(E),采用两血管法建立脑卒中模型,苏木精—伊红染色法(hematoxylin-eosin staining,HE)观察各组大鼠脑皮质病理改变,高倍镜下图像分析软件计数,检测神经元生存密度。结果光镜下可见B组较A组皮质区的细胞排列紊乱,且有坏死软化病灶,神经元出现不同程度的细胞变性,神经元生存密度下降;C、D、E组较B组皮质区细胞坏死减少,变性程度减轻,血管及血管周围间质水肿好转,神经元生存密度增加;E组与C、D组比较,脑皮质区细胞排列较密集,部分锥体细胞胞体突起明显,大部分缺血神经元病理变化明显改善,神经元生存密度明显增加。结论逐瘀通脉胶囊联合针刺可以明显改善大鼠脑皮质区神经细胞病理变化,增加神经元生存密度。
Objective To observe the pathological changes of cerebral cortex in cerebral apoplexy rats after Zhuyu Tongmai capsule combined with acupuncture. Methods 50 healthy male SD rats were randomly divided into five groups： control group （A）, model group （B）, capsule group （C）, acupuncture group （D）, and Zhuyu Tongmai capsule combined with acupuncture group （E）. Two- vessel method was applied to build cerebral apoplexy models, then the changes of rat brain cortical pathology was observed through liE staining and test survival density of the neuron by using counting software at high magnification image. Results Cells in cerebral cortex are more disordered in group B than in group A under light microscope. There were some necrosis and softening lesions, neurons appeared in different degrees of cell degeneration, and survival density of neurons decreased in group B. Compared with group B, necrosis ceils decreased, degeneration degree lessen, blood vessels and interstitial edema around blood vessels turned better, neural survival density increased in group C, D and E. Compared with group C and D, cerebral cortex cells arranged closely, dendrite of some pyramidal cells increased, pathological changes of most ischemic neurons improved, neural density increased significantly in group E. Conclusions Zhuyu Tongmai capsule combined with acupuncture can obviously improve the rats＇ cerebral cortex nerve cell pathological changes and increase the neuronal survival density.
