摘要
目的评价阿奇霉素滴眼液对大鼠和兔眼铜绿假单胞菌性角膜炎的治疗作用。方法采用铜绿假单胞菌分别制备大鼠和兔角膜炎模型,眼部滴入质量分数为1.0%、1.5%、2.0%、2.5%的阿奇霉素滴眼液并与模型组对比,评价其药效学(包括治愈率、清除率、组织病理学的评价)。结果阿奇霉素滴眼液各剂量组对大鼠和兔眼铜绿假单胞菌性角膜炎均具有治疗作用,且呈剂量-效应关系;高剂量(质量分数2.5%)阿奇霉素滴眼液治疗效果优于阳性药(质量分数1.0%眼用凝胶)(P<0.05),中剂量(质量分数2.0%)阿奇霉素滴眼液治疗效果与阳性药相近。结论不同浓度的阿奇霉素滴眼液在大鼠和兔眼角膜炎的治疗试验中均有效,质量分数2.0%和2.5%的阿奇霉素滴眼液疗效更佳。
Objective To evaluate the therapeutic effect of azithromycin eye drops on rat and rabbit keratitis which caused by Pseudomonas aeruginosa. Methods Rat and rabbit keratitis models infected by Pseudo- monas aeruginosa were used, and a single dose of the azithromycin eye drops with the 1.0%, 1.5% ,2.0%, 2. 5 % was instilled into the eyes of the animals. With the saline group as reference, the pharmacokinetic effects of azithromycin eye drops was evaluated by measuring the cure rate, bacterium clearance and his- topathological evaluation after treatment. Results Compared to the saline group, all the rest groups adminis- trated with azithromycin eye drops showed therapeutic effect which was dose-dependent. The high dose (2. 5 % ) group even showed better effect than the positive drug group ( P 〈 0.05 ), and the effect of middle dose(2. 0% ) group was similar to that of the positive drug group. Conclusions All concentrations of azithromy- cin eye drops evaluated in current study are effective in the treatment of rat and rabbit's Pseudomonas aeruginosa keratitis and the therapeutic effect of 2. 0% and 2. 5% azithromycin eye drops is the best among them.
