摘要
建立了大鼠尿液中黄蝶呤、异黄蝶呤、蝶呤-6-羧酸、酪氨酸和色氨酸5种可能的癌症标志物的高效液相色谱-串联质谱(HPLC-MS/MS)分析方法。大鼠尿液除去蛋白后,稀释50倍直接进样分析,采用基质标准曲线法消除基质的影响。上述5种物质的仪器检出限分别为1.6、1.5、2.2、0.9和1.0ng/mL,加标回收率在89.0%~107.0%之间,相对标准偏差(RSD)在0.30%~5.19%之间。
A simple,rapid and sensitive method for the simultaneous determination of five potential cancer biomarkers(xanthopterin,isoxanthopterin,pterin-6-carboxylic acid,tyrosine and tryptophan)in mouse urine by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)has been proposed.After the protein was removed,the mouse urine samples were diluted 50 times and then detected by HPLC-MS/MS directly.The influence of the matrix was eliminated by using the matrix standard curve.The limits of the instrument detection of the five analytes were 1.6,1.5,2.2,0.9and 1.0ng/mL,respectively.The recoveries were in the range of 89.0%-107.0%,and the relative standard deviations between 0.30%and 5.19%.
