低蛋白日粮对育肥猪盲肠代谢产物及菌群的影响 被引量:17

Effects of low protein diets on the metabolites and microbial community in the caecum of fattening pigs

摘要 旨在研究低蛋白添加氨基酸日粮对育肥猪盲肠内代谢产物及食糜与黏膜菌群结构的影响。试验选取12头体重为(60±1)kg的育肥猪,随机分为2组,每组6头,单栏饲养,分别饲喂日粮蛋白水平为10%和16%的低蛋白日粮与正常日粮,试验期50 d。试验结束时,屠宰取盲肠食糜及黏膜样,测定食糜氨氮、微生物蛋白、短链脂肪酸和生物胺的含量,并利用变性梯度凝胶电泳技术及定量PCR技术定性与定量分析盲肠食糜与黏膜微生物的组成及数量变化。结果表明:与对照组相比,低蛋白日粮组中食糜氨氮、微生物蛋白、腐胺和亚精胺的含量显著降低(P<0.05),尸胺和短链脂肪酸的含量无显著变化(P>0.05);食糜微生物组成有显著变化(P<0.05),但总菌、拟杆菌门、厚壁菌门、梭菌XIVa的数量无显著变化(P>0.05),梭菌Ⅳ有降低趋势;黏膜微生物组成及数量均无显著变化(P>0.05)。结果提示,低蛋白日粮能显著降低肠道氮代谢产物,影响食糜微生物组成,对黏膜微生物无显著影响。 The present study was conducted to investigate the effect of low protein diets on the metabolites and microbial community in the caecum of fattening pigs. Twelve fattening pigs with body weights at( 60 ± 1) kg were randomly allocated to 2 groups,which were fed normal diet containing 16% protein and low protein diet containing 10% protein for 50 days,respectively. Each pig was housed individually.Caecal digesta and mucosa were collected for the analysis of ammonia nitrogen,microbial protein,short chain fatty acid,biogenic amines and microbial community and quantity. Results showed that ammonia nitrogen,microbial protein,putrescine and spermidine significantly decreased when fed with low protein diets,and no significant difference was observed with short chain fatty acid and cadaverine. DGGE profiles showed that microbial community in digesta was significantly changed with low protein diets,which was revealed by two separated clusters for each group. Low protein diets had no significant influence on the microbial community in mucosa. Real- time PCR results showed that low protein diets had no significant influence on microbial quantity in both digesta and mucosa except that there was a tendency of decrease with Clostridium cluster IV in digesta. In conclusion,nitrogen related metabolites significantly decreased and microbial community in digesta was affected with low protein diets,while no significant influence was observed with microbial community and quantity in mucosa.
