摘要
用硝酸银染色法对双层平板培养法获得的蛭弧菌进行染色,利用相差显微镜和CCD DP27数码成像系统观察并记录到蛭弧菌形态和侵入大肠杆菌的过程以及蛭弧菌鞭毛的运动状态。同时,通过提高蛭弧菌菌体浓度可观察到蛭弧菌捕捉大肠杆菌的动态过程。该实验让学生更加清晰和直观地观察到菌体的形态及运动方式,激发对微生物实验的热情,促进创新性人才的培养,也为微生物实验教学平台的建设起到一定的推动作用。
Microbiology experimental course plays an important role in microbiology teaching ,focusing on training students’ practical abilities and skills .By silver nitrate staining method ,the Bdellovibrio is stained , which was cultured in double plates .The Bdellovibrio morphology and process of invading E .coli could be clearly observed and recorded using the CCD DP27 digital imaging system of BX43 phase contrast microscope . Furthermore ,the motion of Bdellovibrio flagella are also observed using the system .The dynamic processes of Bdellovibrio capturing E .coli could be observed with the same method when Bdellovibrio is increased the high concentration .Therefore ,students are able to more clear and direct to observe morphology and moment of Bdellovibrio .The experiments can not only stimulate enthusiasm on microbial experiments ,but also promote the cultivation of innovative talents ,and contribute to the construction of experimental teaching platform for the Microbiology as well .
