摘要
目的通过检测非小细胞肺癌患者癌组织和血清中miR-19a、miR-19b的表达水平,探讨其与临床病理参数的关系和作为肿瘤分子标志物的可能性。方法采用实时荧光定量PCR技术检测正常人和非小细胞肺癌患者血清,以及非小细胞肺癌组织和癌旁组织中miR-19a、miR-19b的表达,分析miR-19a、miR-19b的表达与非小细胞肺癌临床病理参数的关系。采用ROC曲线分析miR-19a、miR-19b对非小细胞肺癌的诊断价值。结果 miR-19a、miR-19b在癌组织中的表达量明显高于癌旁正常组织(P<0.05);miR-19a、miR-19b在NSCLC血清中的浓度显著高于对照组。miR-19a的ROC曲线下面积0.989,灵敏度95.1,特异度94.0;miR-19b的ROC曲线下面积0.983,灵敏度93.4,特异度94.0。结论非小细胞肺癌患者癌组织和血清miR-19a、miR-19b高表达,miR-19a、miR-19b有望作为NSCLC诊断及预后评价的检测指标。
Objective To investigate the levels of miR-19 a and miR-19 b expressions in cancer tissues and serum of patients with non-small cell lung cancer(NSCLC),and evaluate the potential of miR-19 a and miR-19 b as biomarkers of NSCLC.Methods Real time-PCR was used to detect the expressions of miR-19 a and miR-19 b in serum of normal people and patients with NSCLC and in cancer tissues and their corresponding non-tumor tissues.The diagnostic value of miR-19 a and miR-19 b for NSCLC was assessed by receiver operator characteristic(ROC)curve.Results The expression levels of serum miR-19 a and miR-19 b were significantly higher in patients with non-small cell lung cancer than in normal people(P〈0.05).The expressions of miR-19 a and miR-19 b in NSCLC were much higher than in the adjacent normal tissues.For miR-19 a the area under ROC curve was 0.989,and the sensitivity and specificity were 95.1 and 94.0.The area under ROC curve for miR-19 b was 0.983,and the sensitivity and specificity were 93.4 and 94.0.Conclusion The high expressions of miR-19 a and miR-19 b are found in cancer tissues and serum from NSCLC patients.Therefore,they may be used as noninvasive biomarkers for diagnosis and prognosis of NSCLC.
