摘要
本文建立了一种水溶液直接浸提结合高效液相色谱法检测红枣中苯甲酸的方法。采用剪切-均质-超声相结合的方式提取红枣中的苯甲酸,比较选择了液相色谱流动相类型与配比,检测了红枣样品中苯甲酸的含量,并进行了加标回收实验。结果显示,以甲醇:水(含0.15%乙酸铵)=5:95时,苯甲酸出峰锋型最佳,保留时间合适,分析速度快。该方法对苯甲酸的提取效率高,回收率在78.8%~105%之间,精密度在4.87%~5.42%之间。方法线性良好(R^2=0.9990),灵敏度高,测定下限为1.5mg/kg,可满足红枣中苯甲酸的检测需求。
In the paper, a water direct extraction combined HPLC method was developed for the determination of benzoic acid in jujube. The combination pretreatment of cut-homogenization-ultrasound was adopted. Different influencing factors such as the proportion of mobile phase were studied. Based on this, real samples determination and recovery experiments were carried out. The result showed that the the mobile phases were methanol and 0.15% ammonium acetate aqueous solution (5:95, v/v). Under these conditions, the peak shape was presented well with an appropriate retention time and rapid measurement. The recoveries were 78.8%~105%, and the RSDs were 4.87%~5.42%. The linearity is good in detection ( R^2=0.9990), and LOQ (S/N =10) was 1.5mg/kg. This method can meet the need for the quantitative detection of benzoic acid in jujube.
