摘要
建立可检测羊乳中掺伪牛乳的间接竞争ELISA方法。通过用牛β-酪蛋白免疫小鼠,制备可特异识别牛乳酪蛋白的单克隆抗体3B9。基于此抗体建立的ELISA的IC50值为92.8 ng/m L牛奶蛋白,理论最低检测限0.35 ng/m L,相对标准偏差小于12%。该方法也适用于检测高温高压处理的乳制品,加之操作简单高效,适合大规模高通量样品筛查及羊乳样本中牛奶蛋白半定量分析。
The aim of the research was to develop an indirect, competitive enzyme linked immunosorbent assay (ELISA) for detecting adulter-ation of bovine milk into goat milk. A monoclonal antibody 3B9 recognizing bovine caseins was selected and characterized after immuniza- tion of mice with bovine β-casein.Once optimized, the half inhibitory concentration (IC50) of mAb3Bg-based ELISA was 92.8 ng/mL of bo- vine protein, with limit of detection of 0.35 ng/mL, the RSD were less than 12%. This assay is also capable of detecting dairy products treat- ed by pasteurization or other severe heat pressure treatment.The method was simple, effective and sensitive, make it suitable for high through-put screening and semi-quantitative analysis of bovine protein in goat milk.
