肺纤方提取物干预肺纤维化模型大鼠微血管内皮细胞黏附及游走迁移作用研究 被引量:5

Influence of Feixian Formula extract on adhesion and migration of microvascular endothelial cell in pulmonary fibrosis rat model

摘要 目的:通过观察肺纤方干预肺间质纤维化大鼠肺微血管内皮细胞体外培养的影响,探讨肺纤方抗肺间质纤维化微血管内皮细胞黏附及游走的机制。方法:从肺纤维化模型组大鼠分离培养肺微血管内皮细胞。将其等量分为空白对照组,氯沙坦(10μg/m L)组,泼尼松(5μg/m L)组,肺纤方大、中、小剂量(100、60、20μg/m L)组。WST-1法检测肺微血管内皮细胞的生长黏附率。Transwell法检测肺微血管内皮细胞的迁移细胞进行计数。结果:与空白对照组、泼尼松组比较,肺纤方大、中剂量和氯沙坦可以显著抑制肺微血管内皮细胞的黏附率,作用48h尤为明显(P<0.01)。与空白对照组、泼尼松组比较,肺纤方大、中剂量和氯沙坦可抑制内皮细胞的迁移(P<0.01)。结论:肺纤方可以显著抑制肺间质纤维化大鼠肺微血管内皮细胞的黏附和游走迁移,从而具有抗肺间质纤维化作用。 Objective: To study the influence of Feixian Formula(FXF) extract on adhesion and migration of pulmonary fibrosis rat microvascular endothelial cell in vitro, and to explore its possible mechanism. Methods: The pulmonary microvascular endothelial cells were isolated and cultured from rat model of pulmonary fibrosis. The cells were divided into six groups: blank control group, losartan(10μg/m L) group, prednisone(5μg/m L) group, FXF high, middle and low dose(100, 60, 20μg/m L) group. The adhesiveness rate of pulmonary microvascular endothelial cells was detected by WST-1, and migration cells from pulmonary microvascular endothelial cells were calculated by transwell method. Results: Compared with blank control group and prednisone group, the adhesiveness rates of pulmonary microvascular endothelial cells in losartan group, FXF high dose group, and FXF middle dose group were all inhibited, especially obvious at 48h(P0.01). Compared with blank control group and prednisone group, the migration cells from pulmonary microvascular endothelial cells in losartan group were inhibited(P0.01). Conclusion: FXF could significantly inhibit the adhesion and migration of pulmonary microvascular endothelial cells in vitro that isolated and cultured from rat model of pulmonary fibrosis, which had an effect of anti-pulmonary fibrosis.
