摘要
目的探讨黄芪含药血清对内皮细胞的保护作用及其可能机制。方法体外培养人脐静脉内皮细胞株(HUVECs)ECV-304,分为阴性对照组、空白对照组、黄芪含药血清干预组。以血管紧张素Ⅱ(AngⅡ)制备血管内皮细胞损伤模型,通过黄芪含药血清进行干预。采用流式细胞仪测定内皮细胞凋亡率,免疫印迹法测定内皮细胞热休克蛋白70(HSP70)的表达。结果 AngⅡ可导致内皮细胞凋亡,不同浓度黄芪含药血清均能够显著降低AngⅡ所致的内皮细胞凋亡率(P<0.05),并可明显促进内皮细胞HSP70表达(P<0.05)。结论黄芪含药血清可抑制AngⅡ诱导的内皮细胞凋亡,促进HSP70表达可能是作用机制之一。
Objective To explore the protective effect of serum contained Astragalus membranaceus on endothelial cell and mechanism. Methods Human umbilical vein endothelial cells (HUVECs) ECV-304 cultured in v itro were divided into the negative control group,the control group and the serum contained Astragalus membranaceus intervention group. Endothelial cell damage mode was established by using angiotensin II (Ang II),and serum contained Astragalus mem-branaceus were used to intervene.Flow cytometry was used to test the apoptosis rate of endothelial cell and im-munoblotting was used to test the expression of heat shock protein 70 (HSP70) in endothelial cell. Results Ang IIcould induce endothelial cell apoptosis.Different concentration of serum contained Astragalus membranaceus were sig-nificantly reduced the apoptosis rate of endothelial cells induced by AngII(P〈0.05),and could obviously promote the en-dothelial cells of HSP70 expression (P〈0.05). Conclusion Serum contained Astragalus membranaceus can inhibit endothe-lial cell apoptosis that is induced by AngII,and promoting the expression of HSP70 may be one of the mechanism.
