宫瘤消胶囊对子宫肌瘤大鼠子宫平滑肌血管内皮生长因子及其受体表达的影响 被引量:13

Effect of GongLiuXiao Capsule on the expressions of vascular endothelial growth factor and its receptors in uterine smooth muscle of rats with hysteromyoma

摘要 目的探讨宫瘤消胶囊对子宫肌瘤大鼠子宫平滑肌血管内皮生长因子(VEGF)及血管内皮生长因子受体1、2(VEGFR1、VEGFR2)表达的影响。方法将60只SD大鼠按随机数字表法分为正常对照组、模型组、桂枝茯苓组以及宫瘤消小、中、大剂量组各10只。除正常对照组外,其余各组采用雌、孕激素负荷法建立大鼠子宫肌瘤模型。以上各组分别给予生理盐水、生理盐水、桂枝茯苓胶囊及宫瘤消胶囊0.54 g/kg、1.35 g/kg、3.38 g/kg连续灌胃4周。透射电镜观察大鼠子宫组织细胞超微结构变化,Western Blot法检测各组大鼠子宫肌层VEGF及其受体VEGFR1、VEGFR2的表达。结果透射电镜结果显示,各剂量宫瘤消胶囊不同程度抑制平滑肌细胞的异常增殖。各组大鼠子宫平滑肌VEGF、VEGFR1、VEGFR2的表达水平比较,差异有统计学意义(P<0.05)。与正常对照组比较,除宫瘤消大剂量组VEGFR2表达差异无统计学意义(P>0.05)外,模型组、桂枝茯苓组及宫瘤消各剂量组VEGF、VEGFR1、VEGFR2的表达水平均显著升高(P<0.05)。宫瘤消小剂量组VEGFR2表达与模型组比较差异无统计学意义(P>0.05),宫瘤消胶囊各剂量组VEGF、VEGFR1、VEGFR2的表达水平均低于模型组(P<0.05)。宫瘤消小剂量组VEGFR2表达与桂枝茯苓组相比无差异外(P>0.05),各剂量组VEGF、VEGFR1、VEGFR2的表达水平均明显低于桂枝茯苓组(P<0.05)。结论宫瘤消胶囊通过抑制子宫肌瘤模型大鼠VEGF及其受体表达,改善子宫肌瘤细胞超微结构,从而达到抑瘤的作用。 Objective To investigate the effect of GongLiuXiao Capsule on the expressions of vascular endothelial growth factor( VEGF) and its receptors( VEGFR1 and VEGFR2) in uterine smooth muscle of rats with hysteromyoma. Methods Sixty SD rats were divided into normal control group,model group,Guizhi Fuling group,low-dosage GongLiuXiao group,middle-dosage GongLiuXiao group and high-dosage GongLiuXiao group by random number table,with 10 rats in each group. Except for normal control group,the rats in the others groups were administrated overload of estrogen and progesterone to establish the rat model of hysteromyoma. The rats in the normal control group,model group,Guizhi Fuling group,low-dosage GongLiuXiao group,middle-dosage GongLiuXiao group and high-dosage GongLiuXiao group were given normal saline,normal saline,Guizhi Fuling Capsule and GongLiuXiao Capsule of 0. 54 g / kg,1. 35 g / kg and 3. 375 g / kg,respectively,by gavage for 4 weeks. The cell ultrastructure of uterine tissues was observed by transmission electron microscope. Western Blot assay was used to detect the expressions of VEGF,VEGFR1 and VEGFR2 in uterine smooth muscle of rats in each group. Results The observation with transmission electron microscope showed that Guizhi Fuling Capsule of various doses could inhibit the abnormal proliferation of smooth muscle cells to different degrees. There was significant differences in the expression levels of VEGF,VEGFR1 and VEGFR2 among six groups( P〈0. 05). Except for VEGFR2 in the high-dosage GongLiuXiao group,the expression levels of VEGF,VEGFR1 and VEGFR2 in the model group,Guizhi Fuling Capsule group and each GongLiuXiao Capsule group were significantly higher compared to those in the normal group( P〈0. 05). There was no significant difference in the expression of VEGFR2 between low-dosage GongLiuXiao Capsule group and model group( P〉0. 05). The expression levels of VEGF,VEGFR1 and VEGFR2 in each GongLiuXiao Capsule group were all lower than those in the model group( P〈0. 05). Except for VEGFR2 in the GongLiuXiao low-dosage group,the expression levels of VEGF,VEGFR1 and VEGFR2 in each GongLiuXiao Capsule group were significantly lower compared to those in the Guizhi Fuling Capsule group( P〈0. 05).Conclusion GongLiuXiao Capsule can inhibit the expressions of VEGF and its receptors in rats with hysteromyoma and improve the ultrastructure of hysteromyoma cells,which plays a role in the inhibition of hysteromyoma.
