摘要
目的探讨姜黄素对DSS诱导溃疡性结肠炎小鼠T淋巴细胞亚群及BTLA表达的影响。方法采用葡聚糖硫酸钠法(DSS)制备UC小鼠模型,小鼠随机分为4组:对照组、DSS模型组、地塞米松组、姜黄素组,通过腹腔注射药物作用,应用流式细胞仪检测T淋巴细胞亚群和细胞因子表达的百分率。结果姜黄素组与DSS模型组比较,小鼠外周血CD4+细胞数明显升高,CD8+细胞数明显降低,CD4+/CD8+比值明显升高(Р<0.01);小鼠脾脏Th1+细胞、Tc1+细胞比例和Th1+/Th2+比值、Tc1+/Tc2+比值明显降低,而Th2+细胞明显升高(P<0.05);小鼠外周血、脾脏CD4+T细胞上BLTA+的表达明显增高(P<0.01)。结论姜黄素对DSS诱导UC小鼠CD4+T细胞亚群及BTLA表达显著升高,可能是通过调节CD4+/CD8+T淋巴细胞亚群间的平衡,抑制Th1型/Tc1型细胞分化,促使CD4 T淋巴细胞表面BTLA表达上调,降低免疫反应,发挥抗炎及治疗UC作用。
Objective To investigate the effect of curcumin on T lymphocyte subsets and BTLA expression in mice with ulcerative colitis induced by DSS. Methods 5% Dextran sulfate sodium( DSS) was used for the establishment of ulcerative colitis( UC) rat model,and then UC rat model was divided into four groups: control group,DSS model group,dexamethasone group and curcumin group( 30 mg / kg). The proportion of T lymphocyte subsets in peripheral blood and expression of cytokines were calculated by flow cytometry.Results In this study,we observed an increase in CD4 + T cell was associated with treatment of curcumin( P〈0. 01). Contrary to CD4 + T cell,the number of CD8- T cell and ratio of CD4 + / CD8 + in curcumin group was lower than model group( P〈0. 01). The loss of Th1 +,Tc1 +,Th1 + / Th2 + and Tc1 + / Tc2 + was also observed in curcumin group( P〈0. 05). The high level of BTLA expression was showed in curcumin group( P〈0. 01). Conclusions Curcumin shows obvious effect on CD4 + T cell subsets and BTLA expression,and the therapeutic mechanism might be related with the balance of CD4 + / CD8 + T cell,the inhibition of Th1 / Tc1 differentiation and up- regulation of BTLA expression.
