摘要
目的观察分子结构乙酰化修饰对枸杞多糖(Lycium barbarum L.polysaccharide,LBP)体内抗氧化、抗肿瘤活性的影响。方法通过分子结构乙酰化修饰得到枸杞多糖乙酰化衍生物LBPa;将昆明种小鼠随机分成空白对照组,阳性对照组(维生素C),LBP高、低剂量组及LBPa高、低剂量组,测定小鼠血清和肝脏丙二醛(malondialdehyde,MDA)、肝脏总抗氧化能力(total antioxidant capacity,T-AOC)和过氧化氢酶(catalase,CAT)。复制H22荷瘤小鼠模型,灌胃给药LBP,考察LBPa和LBP对肿瘤细胞抑制率、脾脏指数、胸腺指数及血清肿瘤坏死因子α(tumor necrosis factorα,TNF-α)和白介素-2(interleukin,IL-2)含量的影响。结果 LBPa可以显著提高正常小鼠肝组织CAT活力及T-AOC,降低小鼠血清及肝脏MDA含量,其抗氧化效果优于维生素C及LBP,LBPa及LBP抗氧化活性均与剂量具有正相关性。LBPa可显著抑制H22肿瘤细胞增殖,提高免疫器官指数,提高血清IL-2及TNF-α含量;除IL-2含量外,对其他指标的影响均有剂量依赖性;LBPa抗肿瘤活性明显优于LBP。结论乙酰化修饰可显著提高LBP的抗氧化及抗肿瘤活性。
Objective To observe the influence of acetylation on the in vivo antioxidant and antitumor activities of Lycium barbarum L. polysaccharide (LBP). Methods LBPa, the acetyl derivative of LBP, was obtained through acetylation. Kunming mice were randomly divided into blank control group, positive control group (vitamin C), low- and high-dose LBP groups, and low- and high-dose LBPa groups, and serum and liver malondialdehyde (MDA), total antioxidant capacity (T-AOC) of the liver, and catalase (CAT) were determined. A H22 tumor-bearing mouse model was established, and these mice were given LBP by gavage. The influence of LBPa and LBP on the inhibition rate of tumor cells, spleen index, thymus index, and serum levels of tumor necrosis factor-α (TNF-α) and interleukin-2 (IL-2) was observed. Results LBPa significantly improved liver CAT activity and T-AOC in normal mice and reduced the content of serum and liver MDA, achieving a better antioxidant effect than vitamin C and LBP; the antioxidant activity of LBP and LBPa was positively correlated with their doses. LBPa significantly inhibited the proliferation of H22 tumor cells, improved the immune organ index, and increased the serum levels of IL-2 and TNF-α; the influence on all indices except IL-2 was dose-dependent; LBPa had a better antitumor activity than LBP. Conclusion Acetylation can significantly improve the antioxidant and antitumor activities of LBP.
