Optimization of Culture Conditions Using Response Surface Methodology for Biomass Production of Streptomyces ahygroscopicus 001 Strain

摘要 为提高不吸水链霉菌梧州专化型001菌株发酵的生物量,本研究采用响应面法对该菌株的液体发酵条件进行优化。在单因素试验的基础上,利用Plackett-Burman(P-B)试验设计证明可溶性淀粉(X1)、初始p H值(X4)以及温度(X5)3种因素对不吸水链霉菌生物量的积累存在着较为显著的影响。在响应面分析(response Surface Methodology,RSM)中,X1、二次项X42和X52均能显著影响该菌株生物量的积累。优化的最佳发酵条件组成为:可溶性淀粉34.66g·L-1、p H值7.62、温度28.29℃,此时不吸水链霉菌菌体生物量达0.0137g·L-1。验证试验结果表明:在优化后的培养条件下菌体生物量要比优化前提高73.1%。 Streptomyces ahygroscopicus neosub. sp. wuzhouensis 001 had a broad antipathogen spectrum. In order to obtain the highest biomass of Streptomyces ahygroscopicus, response surface methodology was used to optimize ferment variables of the strain. Three variables including soluble starch(X1), initial p H(X4) and temperature(X5) had significant effects on the production of biomass were selected based on the Plackett-Burman(PB)design. During the analysis of response surface, X1, quadratic terms X42 and X52significantly influenced the biomass production. The optimized conditions for promoting biomass production were as follows: soluble starch, 34.66g·L-1; p H 7.62 and temperature, 28.29℃. Under these conditions, the biomass increased to 0.0137g·L-1.Verification test demonstrated that optimized conditions improved Streptomyces ahygroscopicus 001 biomass by 73.1 % comparing with un-optimized conditions treatment.
