摘要
为充分利用废弃物资源,以废弃平菇栽培料为原料,经浸提正交优化、硫酸铵分级沉淀、透析、双水相萃取等方法,提取、加工高纯度纤维素酶制剂。结果表明:以0.2 mol/L p H 5.5的乙酸-乙酸钠为缓冲液,料液比为1∶4,温度15℃,浸提1~2 h,再利用质量分数均为20%的PEG6000-硫酸铵双水相体系进行萃取,纯化后的纤维素酶比活力为19 705.60 U/mg,纯化倍数为5.34,回收率为60.9%。
In order to take full use of waste resources,the waste Pleurotus ostreatus compost was selected to extract and process high-purity cellulase by orthogonal optimization,ammonium sulfate precipitation,dialysis and aqueous two-phase extraction method. Results showed that the specific activity,purification fold and recovery rate of the resultant enzyme was 19 705.60 U/mg,5.34 and 60.9%,respectively,after using 0.2 mol/L acetic acid-sodium acetate as buffer solution(p H 5.5),ratio of material to liquid at 1∶4,immersing for 1-2 h at 15℃ and extraction by aqueous twophase system composed of PEG6000 and ammonium sulfate with mass fraction of 20%.
