摘要
目的探讨五味子乙素(Sch B)对苯并芘(Bap)所致不育模型小鼠精液质量降低的保护作用及其机制。方法选取性成熟雄性美国癌症研究所小鼠60只,随机分为对照组、Bap组和Sch B+Bap组,每组20只。Bap用橄榄油配成0.2 g·L-1混悬液,Sch B用9 g·L-1氯化钠注射液配成20 g·L-1混悬液。Bap组小鼠给予Bap 2 mg·kg-1·d-1灌胃,Sch B+Bap组小鼠给予Bap(2 mg·kg-1·d-1)及Sch B(200 mg·kg-1·d-1)灌胃,对照组小鼠给予相同剂量的9 g·L-1氯化钠注射液与橄榄油灌胃,30 d后处死小鼠取出睾丸组织,观察各组小鼠的精子密度、活率及动态参数曲线运动速度(VCL)、直线运动速度(VSL)、平均路径速度(VAP)、平均移动角度(MAD)、精子头侧摆幅度(ALH)、鞭打频率(BCF)、直线性(LIN)、摆动性(WOB)、向前性(STR),并测定睾丸组织细胞色素P450(CYP450)、谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)活性及凋亡相关基因Bax、Bcl-2 mRNA表达水平。结果与对照组小鼠比较,Bap组小鼠的精子密度、活率以及a级精子与b级精子活率均显著降低(P<0.05);Sch B+Bap组小鼠的精子密度、活率、a级精子与b级精子活率显著高于Bap组(P<0.05)。Sch B+Bap组小鼠精子密度低于对照组(P<0.05),但2组小鼠间精子活率比较差异无统计学意义(P>0.05)。Bap组小鼠的各项精子动态参数均低于对照组(P<0.05)。Sch B+Bap组小鼠的各项精子动态参数均高于Bap组(P<0.05)。Sch B+Bap组小鼠精子的ALH、STR、BCF均显著低于对照组(P<0.05),VCL、VSL、VAP、LIN、MAD、WOB与对照组小鼠比较差异无统计学意义(P>0.05)。与对照组小鼠比较,Bap组、Sch B+Bap组小鼠睾丸组织中CYP450活性显著增高(P<0.05),GSH-PX、SOD活性显著降低(P<0.05)。Sch B+Bap组小鼠睾丸组织中CYP450活性显著低于Bap组(P<0.05),GSH-PX、SOD活性显著高于Bap组(P<0.05)。与对照组小鼠比较,Bap组、Sch B+Bap组小鼠睾丸组织中Bcl-2 mRNA水平显著降低(P<0.05),Bax mRNA水平及Bax/Bcl-2显著增高(P<0.05)。Sch B+Bap组小鼠睾丸组织中Bcl-2 mRNA水平显著高于Bap组(P<0.05),Bax mRNA水平及Bax/Bcl-2显著低于Bap组(P<0.05)。结论五味子乙素可以改善苯并芘导致的小鼠精液质量降低,其作用可能是通过提高抗氧化酶活力及调节凋亡相关因子而实现。
Objective To investigate the role of schisandrin B( Sch B) on sperm quality in mice model of infertility induced by benzo-( α)-pyrene( Bap) and its mechanism. Methods Sixty viripotent male Institute of Cancer Research mice were randomly divided into control group,Bap group and Sch B + Bap group,each group included 20 mice. Bap suspension was prepared with olive oil( concentration 0. 2 g·L- 1),Sch B suspension was prepared with 9 g·L- 1sodium chloride injection( concentration 20 g·L- 1). Male mice were lavaged with 9 g·L- 1sodium chloride injection and olive oil in control group.The mice were lavaged with Ba P( 2 mg·kg- 1·d- 1) in Bap group,and the mice were lavaged with Bap( 2 mg·kg- 1·d- 1)and Sch B( 200 mg·kg- 1·d- 1) in Bap + Sch B group. After 30 days,all the mice were executed and the testicle were extracted to detected the sperm density,motility rate,different dynamic parameters[VCL,VSL,VAP,mean angular displacement( MAD),ALH,beat cross frequency( BCF),linearity( LIN),wobble( WOB),straightness( STR) ]. The activity of cytochrome 450( CYP450),glutathione peroxidase( GSH-PX),superoxide dismutase( SOD) and Bax,Bcl-2 mRNA level in the testicle were also examined. Results Compared with control group,sperm density,motility rate of mice in Bap group decreased significantly( P〈0. 05),which in Bap + Sch B group increased significantly than those in Bap group( P〈0. 05). The sperm count in Sch B + Bap group was significantly decreased compared with control group( P〈0. 05),and there was no significant difference of motility rate between the two groups( P〈0. 05). The dynamic parameters of mice in Bap group were significantly lower than those in control group,which in Sch B + Bap group were significantly higher than those in Bap group( P〈0. 05).ALH,STR,BCF of mice in Bap + Sch B group were significantly lower than those in control group( P〈0. 05),but VCL,VSL,VAP,LIN,MAD,WOB had no significant difference between control group and Bap + Sch B group( P〈0. 05). Compared with control group,the activity of cytochrome P450 was increased significantly while the activity of GSH-PX,SOD decreased significantly in Bap group and Bap + Sch B group( P〉0. 05). Compared with Bap group,the activity of cytochrome P450 was significantly lower while the activity of GSH-PX,SOD were significantly higher in Bap + Sch B group( P〈0. 05). Compared with control group,the expression of Bcl-2 mRNA level increased significantly while the expression of Bax mRNA level and Bax /Bcl-2 increased significantly in Bap group and Bap + Sch B group( P〈0. 05). Compared with Bap group,the expression of Bcl-2 mRNA level increased significantly while the expression of Bax mRNA level and Bax / Bcl-2 increased significantly in Bap + Sch B group( P〈0. 05). Conclusion Sch B can improve sperm quality of infertility mice induced by Bap,this effect may be related to antioxidation and inhibition of cell apoptosis.
