摘要
目的:分析有窦型与无窦型根尖周炎感染根管内菌群结构的差异。方法:采集17例原发性根尖周炎的根管样本,其中有窦型9例、无窦型8例。分别经DNA抽提和16S r RNA基因V3区PCR扩增后,利用变性梯度凝胶电泳(DGGE)检测两组样本的微生物群落结构;然后分别采用聚类分析、主成分分析(PCA)和偏最小二乘法(PLS)对两组样本的菌群结构进行比较,并对两组间有显著差异的相关区别条带进行割胶测序。结果:聚类分析、PCA、PLS显示,有窦型与无窦型样本的菌群结构存在差异,共有5条区别条带的检出水平在两组间显著不同(P<0.05)。测序结果提示,Dialister invisus、Bacteroidetes clone_X083、Pseudoramibacter alactolyticus与有窦型根尖周炎相关,而嗜糖普雷沃菌和纤细弯曲菌则与无窦型根尖周炎相关。结论:有窦型和无窦型根尖周炎感染根管内的微生物群落结构存在差异,Dialister invisus、Bacteroidetes clone_X083、Pseudoramibacter alactolyticus可能与窦道形成有关。
AIM: To compare the microbiota in the infected root canals of apical periodontitis with and with- out sinus tract. METHODS: 17 root canal samples were collected from patients of primary endodontic infections, in- cluding 9 cases with and 8 without sinus tract. DNA was extracted and amplified by PCR based on the V3 region of 16S rRNA gene. The microbial profiles of the samples were analyzed by denatured gradient gel electrophoresis (DGGE). Cluster analysis, principle component analysis (PCA) and partial least square (PLS) were used to reveal the differ- ence of the microbiota between the samples with and without sinus tract, and bands with significantly different detection level between the 2 groups were excised for sequencing. RESULTS: 5 bands were found with significant different de- tection level between the 2 groups ( P 〈 0.05 ). Sequencing results indicated that Dialister invisus, Bacteroidetes clone_ X083 and Pseudoramibacter alactolyticus were associated with the samples with sinus tract, while Prevotella multisac- charivorax and Campylobacter gracilis were associated with those without sinus tract. CONCLUSION: The microbial community of apical periodontitis with sinus tract is different from those without sinus tract. Dialister invisus, Bacte- roidetes c/one_X083 and Pseudoramibacter alactolyticus are suggested to be related to the sinus tract formation.
