甘肃部分玉米自交系SSR遗传多样性分析 被引量:4

摘要 分析甘肃省部分骨干玉米自交系的遗传多样性并划分群体结构。利用全自动DNA分析仪荧光SSR-PCR技术和66对核心SSR引物研究了148份玉米(Zea mays L.)自交系的遗传多样性,并分别使用模型结构聚类和遗传距离聚类法进行群体结构分析。在148份自交系中,66对SSR标记共检出279个等位变异,每对引物检测到等位基因2-8个,平均4.2273个。多态性信息量(PIC)和标记指数(MI)的平均值分别为0.4879和2.1808。模型结构聚类将148份自交系分为SS群和NSS群;遗传距离聚类分为5个类群,根据常规测验种自交系可将5个类群归并为SS群和NSS群。2种聚类方法所得结果与材料的系谱信息基本一致。甘肃省部分玉米自交系在育种实践中正在向2个相对独立的优势类群转化,2个类群之间的遗传多样性水平存在一定的差异。 The study aims to analyze the genetic diversity and divide the population structure of some maizeinbred lines in Gansu Province. Genetic diversity of 148 maize inbred lines was analyzed with 66 pairs of coreSSR primers and fluorescent SSR-PCR technology in automatic DNA analyzer. The population structure wasanalyzed by distance-based cluster and model-based cluster methods. Totally in the 148 lines, 279 alleleswere detected with an average of 4.2273, ranged from 2 to 8 per locus with 66 pairs of SSR primers. Theaverage polymorphism information content(PIC) and marker index(MI) were 0.4879 and 2.1808, respectively.All the 148 inbred lines were grouped into NSS group and SS group by model-based cluster method, and weredivided into five groups by distance-based cluster method. These 5 groups could be incorporated into SS groupand NSS group according to the commonly used test inbred lines. Results showed that the clustering groupswere consistent with the basic materials of pedigree information. In conclusion, part of maize inbred lines inGansu Province were transforming into two relatively independent groups in breeding practice, the geneticdiversity level between the two groups had certain differences.
