摘要
为了解抗氧化剂减缓长期继代蝴蝶兰类原球茎衰老的机理,本实验在培养基添加抗氧化剂100 mg/L半胱氨酸和8 g/L甘露醇继代类原球茎24个月,分析其相关氧化指标及DNA稳定性。结果表明,类原球茎培养期间处理组总活性氧水平变化趋势与对照组不同,处理组比对照组低,对照组总活性氧水平消长趋势与超氧阴离子(O_2^-)生成速率较为一致,处理组则与羟自由基(·OH)相对含量的消长趋势较一致。继代期间丙二醛含量变化趋势与总活性氧水平变化趋势相似,处理组比对照组低。类原球茎DNA的SRAP分析结果表明,继代培养期间处理组与对照组间均未发生明显的DNA差异。类原球茎DNA甲基化程度变化趋势与总活性氧水平变化大体相同,中后期处理组比对照组低。
To understand the mechanism that antioxidants slowed down the aging of Phalaenopsis PLB through long-term subculture, PLB were subcultured for 24 months in the culture supplemented with antioxidants (100 mg/L cysteine and 8 g/L mannitol), and the related oxidant indexes and DNA stability of PLB were analyzed in the experiment. The results showed that during subcuhuring of PLB the variation tendency of levels of total reactive oxygen species (T-ROS) was not consistent between the control and the treatment. The T-ROS levels of the treatment were lower than that of the control. During PLB subculturing, the variation tendency of the levels of T- ROS in the control was consistent with that of the growth rate of superoxide anion (O2^·-), but it accorded with the relative quantity of hydroxyl radical (.OH) in the treatment. The changes of malondialdehyde content were generally consistent with the levels of T-ROS during subcuhuring, the contents were lower in the treatment than that in the control. The result of SRAP analysis on genome DNA indicated the polymorphic difference did not show clearly in the PLB throughout subculturing, as well as between the control and the treatment. The variation tendencies of levels of DNA methylation of PLB and the levels of T-ROS were about the same. However, the levels in the treatment were lower than that in the control at middle-late stage.
