摘要
目的观察干性转录因子KLF4及Ki-67、E-cadherin在宫颈鳞状细胞癌组织中的表达,探讨三者表达的关系及其意义。方法采用免疫组化检测KLF4、Ki-67和E-cadherin在高-中分化鳞状细胞癌(28例)、低分化鳞状细胞癌(52例)和慢性宫颈炎(26例)组织中的表达,并统计分析三者表达的关系及KLF4在宫颈鳞状细胞癌中的表达与临床病理特征的关系。结果 KLF4在慢性宫颈炎中均呈阳性表达,而宫颈鳞状细胞癌中有不同程度下调,在高及中分化宫颈鳞状细胞癌中阳性率为85.7%(24/28),低分化宫颈鳞状细胞癌中为59.6%(31/52),两者比较差异显著(P<0.01)。宫颈鳞状细胞癌中,KLF4表达与年龄、肿瘤大小、淋巴结转移、远处转移及肿瘤分期无关,而与肿瘤分级有关(P<0.05)。KLF4表达与增殖指数Ki-67呈负相关(P<0.05),而与E-cadherin呈正相关(P<0.01)。结论 KLF4在宫颈鳞状细胞癌中与细胞增殖和迁移相关,KLF4可能在宫颈鳞状细胞癌发生、发展中起重要作用。
Purpose To investigate the expression of stem cell specific transcription factor KLF4 and Ki-67, E-cadherin in tissues of cervical squamous cell carcinoma and explore the correlation among these three factors and their significance. Methods Immunohistochemical detection of KLF4, Ki-67 and E-cadherin in high-moderately differentiated squamous cell carcinoma (28 cases) , poorly differentiated squamous cell carcinoma (52 cases) and chronic cervicitis (26cases) tissues, and statistical analysis of the expression of KLF4 in cervical squamous cell carcinoma tissues and it' s relationship with clinicopathological features in squamous cell carcinoma. Results KLF4 expressed in all chronic cervicitis tissue. KLF4 downregulated in cervical squamous cell carcinoma as compared to chronic cervicitis. The positive rate of KLF4 is 85.71% (24 out of 28) in high-moderate differentiated cervical squamous cell carcinoma, 59. 62% (31/52) in low differentiated cervical squamous cell carcinoma and the difference between them was statistically significant ( P 〈 0. 01 ). KLF4 expression had no correlation with age, tumor size, lymphatic metastasis, distant metastasis and tumor staging in cervical squamous cell carcinoma, but do with tumor grading. KLF4 expression was negatively correlated with Ki-67 proliferation index ( P 〈 0. 05 ), but positively correlated with E-cadherin ( P 〈 O. 01 ). Conclusions KLF4 expression is assoicated cell proliferation and migration markers and maybe play a potential role development and progression in cervical squamous ceil carcinoma.
