自来水管网生物膜中的细菌检测及卫生学评价

Detection of bacteria and hygienic evaluation of biofilms in tap water

摘要 目的检测分析自来水管网生物膜中细菌的类群及其可能对人类健康的威胁,为自来水管网生物膜的卫生学评价提供参考。方法截取某市政自来水管网实际管段,冲刷收集管壁生物膜,共采集10个生物膜样品,参照GB/T5750.12—2006检验方法测定细菌总数和大肠菌群,参照2010年版中国药典关于纯化水的微生物检测方法测定贫养细菌,使用流式细胞法检测活菌数目,用志贺菌显色培养基、沙门菌显色培养基、BCYE培养基和TCBS培养基对志贺菌、沙门菌、军团菌和霍乱弧菌分离鉴定并计数。结果 10个生物膜样品中,营养琼脂法检测细菌数目为101cfu/cm2~104cfu/cm^2,R2A琼脂培养法细菌计数为102cfu/cm^2~106cfu/cm^2,流式细胞法检测的活菌数目为104cfu/cm^2~108cfu/cm^2。大肠菌群检出率达80.0%,志贺菌检出率为60.0%,沙门菌检出率为50.0%,未检出军团菌和霍乱弧菌。结论自来水管网生物膜存在大量细菌,自来水管网生物膜对人体健康存在潜在威胁。 Objective To investigate bacteria species and their potential threat to human health in biofilm in tap water distribution system. Methods Cut a part of the actual pipe section of the municipal water pipe network of some city and scour the biofilm of the pipe wall. 10 biofilm samples were collected. The total count of bacteria and coliform bacteria were detected according to GB / T 5750. 12—2006,while the oligotrophic bacteria was detected by microbial detection method for purified water in2010 edition of Chinese Pharmacopoeia,using flow cytometry to detect the number of live bacterium. Bacteria Shigella,Salmonella,Legionella and Vibrio cholera were isolated using Shigiella chromogenic medium,Salmonella chromogenic medium,BCYE and TCBS respectively,then identified according their biochemical reaction characteristics. Results 10 biofilm samples were detected by nutrient agar method,the total count of bacteria of 13 samples ranged from 101 cfu / cm2- 104 cfu / cm2; detected by R2 A agar culture method,the count of the bacteria was among 102 cfu / cm2- 106 cfu / cm2; detected by flow cytometry,the count of live bacterium was among 104 cfu / cm2- 108 cfu / cm2. The detection rate of Coliforms bacteria was 80. 0%,Shigella was 60. 0%,and Salmonella was 50. 0%. No Legionella and Vibrio cholera were detected out. Conclusion There was a large amount of bacteria in biofilm collected from tap water distribution system,which will threaten human health potentially.
