辛伐他汀对血管紧张素Ⅱ诱导心肌细胞损伤模型大鼠心肌细胞ACE2的影响 被引量:1

Effects of Simvastatin on ACE2 Cells in Rats with Angiotensin Ⅱ Induced Myocardial Injury

摘要 目的观察辛伐他汀(SIM)对血管紧张素Ⅱ(AngⅡ)诱导心肌损伤模型大鼠心肌细胞ACE2的影响。方法制备AngⅡ诱导心肌细胞损伤大鼠模型,随机分为空白对照组、AngⅡ组、小剂量他汀组+AngⅡ组、中剂量他汀组+AngⅡ组、高剂量他汀组+AngⅡ组。应用Western blot、PCR技术检测各组大鼠心肌细胞ACE2蛋白和基因表达。结果与空白对照组比较,心肌损伤模型组(Ang II组)ACE2蛋白和基因表达下降(0.21比0.74,0.333±0.122比1.000±0.149),两组差异有统计学意义(P<0.05);与AngⅡ组比较,小剂量他汀组ACE2蛋白(0.39)和基因表达(0.399±0.054)略升高,但差异无统计学意义(P>0.05);中剂量他汀组ACE2蛋白(0.45)和基因表达(0.644±0.135)、高剂量他汀组ACE2蛋白(0.61)和基因表达(0.952±0.090)与AngⅡ组比较差异均有统计学意义(P<0.05),且ACE2蛋白和基因表达随着辛伐他汀浓度的升高而上升。结论 AngⅡ刺激心肌细胞致ACE2蛋白、ACE2基因表达下降,而辛伐他汀可抑制该作用,使受AngⅡ损伤的心肌细胞ACE2蛋白、ACE2基因表达升高,其作用随药物浓度升高而增加。 Objective To investigate the effects of simvastatin on angiotensin converting enzyme 2(ACE2)cells in the process of angiotensin II(Ang II)induced myocardial injury in rats.Methods Myocardial cells were cultured in vitro and pretreated with Ang II,then divided into Ang II group,low-dose SIM +Ang II group,mid-dose SIM+Ang II group,and high-dose SIM+Ang II group.Normal myocardial cells served as control group.Western Blot and PCR technique were used to detect expressions of ACE2 protein and gene in all groups.Results Compared with control group the ACE2 protein and gene expressions in Ang II group decreased(0.21 vs 0.74,0.333±0.122 vs1.000±0.149;P〈0.05).Compared with Ang II group,the expressions of ACE2 protein and gene in mid-dose SIM+Ang II group(0.45,0.644±0.135)and high-dose SIM+Ang II group(0.61,0.952±0.090)significantly increased(all P〈0.05),but those in low-dose SIM +Ang II group slightly increased in the absence of significant difference(0.39,0.399±0.054;P〉0.05).The expressions of ACE2 protein and gene elevated with the increase of simvastatin concentration.Conclusion Ang II decreases the expression of ACE2 protein and gene on myocardial cells.Simvastatin can inhibit this effect,by increasing the expression of ACE2 protein and gene on Ang II-injued myocardial cells.This inhibition effect is in dose-dependent manner.
