摘要
目的通过建立兔膝关节骨性关节炎(OA)模型,探讨白细胞介素(IL)-1β、基质金属蛋白酶(MMP)-9在OA滑膜中的表达及其意义。方法60只新西兰大白兔随机分为6组,A1、A2、A3组为模型组,B1、B2、B3组为对照组。A1、A2、A3组用1.6%木瓜蛋白酶膝关节腔内注射,建立兔膝关节OA模型,分别在2、4、6周后处死,观察大体标本形态学变化及标本滑膜中IL-1β和MMP-9的表达情况。结果(1)实验组4周和8周软骨细胞IL-1β和MMP-9细胞分数明显高于对照组,差异有统计学意义(P〈0.05或P〈0.01),实验组软骨细胞IL-1β和MMP-9细胞分数4周较8周组明显增高,差异有统计学意义(P〈0.05)。(2)酶联免疫吸附试验(ELISA)法检测IL-1β、MMP-9的表达:A1组〉B1组、A2组〉B2组、A3组〉B3组(P〈0.05),模型组中Al组〈A2组、A2组〈A3组(P〈0.05)。(3)Westernblot法检测IL-1β蛋白的相对表达水平:模型组滑膜中的IL-1β的相对表达量均高于对照组,A1组(0.80±0.05)〉B1组(0.40±0.02)、A2组(1.08±0.06)〉B2组(0.51±0.03)、A3组(1.26±0.08)〉B3组(0.61±0.03,P〈0.01),且随实验进展时间的延长而增加。A1组〈A2组,A2组〈A3组(P〈0.05)。结论IL-1β和MMP-9的差异表达与OA退变的发展密切相关。
Objective To investigate the expression and significance of matrix metalloprotein -9 (MMP -9) and interleukin (IL) - 1β in the synovial membrane of osteoarthritis (OA) model. Methods Sixty New Zealand white rabbits were randomly divided into 6 groups: AI, A2, A3 groups as model groups, and Bl, B2, B3 groups as the control groups. In A1, A2 and A3 groups, 1.6% papain was injec- ted into the articular cavity to establish the OA model of the knee joints, and the animals were sacrificed at 2nd, 4th and 6th week. The expression of MMP - 9 and IL - 1β in the specimens was observed. Results ( 1 ) At 4th week and 8th week, the MMP - 9 and IL - 113 expression levels in the model groups were sig- nificantly higher than those in the control groups ( P 〈 0. 05 and P 〈 0.01 ). At 4th week, the MMP - 9 and IL - 1β levels in the model groups were significantly higher than those at 8th week (P 〈 0. 05 ). (2) The expression levels of IL - 1β and MMP -9 in group A1 were higher than in group B1, group A2 higher than in group B2, group A3 higher than in group B3 ( P 〈 0. 05 ), group A1 lower than in group A2 and group A2 lower than in group A3 .(P 〈0. 05). (3) For Western blotting, the content of IL - 1β in model groups was higher than in control groups : group A1 (0. 80 ± 0. 05 ) higher than in group B1 (0. 02 ± 0. 40 ), group A2 ( 1.08 ± 0. 06) higher than in group B2 (0.06 ± 1.08 ), group A3 (0.03 ± 0. 08 ) high- er than in group B3 (0. 61 ±0. 03, P〈0. 01), and increased with the time in the model groups: group A1 lower than in group A2, and group A2 lower than in group A3 (P 〈 0. 05 ). Conclusion The differential expression of IL - 113 and MMP -9 is closely related to the development of OA degeneration.
