摘要
目的:探讨保存温度和时间对肝功能血清酶活性的影响,旨在保证高质量的血清样本用于生化检测。方法:采集15只BALB/c小鼠血清,部分血清标本立即进行检测,部分标本贮存在不同条件下(室温24~26℃下保存1d,+4℃保存1d和2d,-20℃保存1个月),然后再检测。所有标本均采用酶联免疫吸附实验(elisa法)检测丙二醛(MDA)、超氧化物歧化酶(SOD)、甘油三酯(TG)、谷胱甘肽过氧化物酶(GSH-Px)和分泌型磷脂酶A_2(sPLA_2)酶活性的变化,并对结果进行统计学分析。结果:(1)血清在室温贮存1d后检测酶的含量是不准确的。(2)血清在+4℃贮存1d,-20℃贮存1个月仍可同时检测SOD、TG、sPLA_2、MDA和GSH-Px的含量。(3)血清在+4℃贮存2d可同时检测SOD和sPLA_2的含量。结论:进行大批量肝功能血清酶检测时,应考虑存储时间和温度等因素对血清样本的影响,以确保酶活性数据分析的客观性和真实性。
Objective:To explore the effect of preservation temperature and time on the activity of serum enzymes in liver function and ensure the high quality of serum samples for biochemical test.Methods:The BALB/c serum samples were collected from 15 mice.Some specimens were immediately tested,and the remaining specimens were stored under different conditions(room temperature 24~26 ℃for a day,+4 ℃for a day and two day,-20 ℃for a month).All specimens were detected the changes of malondialdehyde(MDA),superoxide dismutase(SOD),triglyceride(TG),glutathione peroxidase(GSH-Px)and secretory phospholipase A_2(sPLA_2)by Elisa method.The results were statistically analyzed.Results:(1)Serum enzyme levels were not accurate when stored at room temperature for a day.(2)Enzyme content of SOD,TG,sPLA_2,MDA and GSH-Px could be detected simultaneously at+4 ℃ for a day and-20 ℃ for a month.(3)Enzyme content of SOD and sPLA_2 could be detected simultaneously at+4℃for two day.Conclusion:To ensure the objectivity and authenticity of the data analysis,the effect of storage time and temperature should be taken into consideration when large quantities of serum enzyme is detected in liver function.
